Basic Information
Tvardi Therapeutics is a clinical-stage private biotechnology company focused on developing small-molecule inhibitors targeting STAT3. STAT3 is a key regulatory protein at the intersection of multiple signaling pathways and plays a crucial role in cancer cell survival and immune evasion. The company’s lead therapy, TTI-101, is an orally administered small-molecule STAT3 inhibitor that has demonstrated anticancer activity in animal models of head and neck cancer, lung cancer, breast cancer, and liver cancer. It is currently in Phase 1 clinical trials for the treatment of patients with advanced solid tumors. To date, it has exhibited a favorable safety profile and clinical activity. STAT3 is also a critical mediator of chronic inflammation and fibrosis. The company’s second-generation STAT3 inhibitors will be used to treat fibrotic diseases. They have shown the ability to reverse the fibrotic process and restore normal tissue function in animal models of idiopathic pulmonary fibrosis, non-alcoholic steatohepatitis, and scleroderma.
