Rapafusyn's mission is to develop transformative drugs to meet substantial unmet medical needs and improve patient care. The platform leverages non-degrading macrolide molecular glues (RapaGlues™) to address challenging therapeutic targets. Rapafusyn has built DNA-encoded libraries (DEL) and an array library of RapaGlues™, which can form new PPIs and inhibit the native activity of proteins. These compounds are rationally designed and engineered to bind with FKBP and form ternary complexes with disease target proteins, providing a promising pathway for modulating complex protein-protein interactions. RapaGlues™ have proven effective in delivering innovative chemical starting points for challenging drug targets. Their modular structure allows for SAR and subsequent optimization of efficacy, selectivity, and physicochemical properties, thereby accelerating drug discovery and development. RapaGlues™ often exhibit cell permeability directly from the starting point, enhancing their therapeutic development potential.
