Imbria is a privately held clinical-stage company developing novel therapies for patients with life-altering cardiometabolic diseases. Our clinical pipeline focuses on restoring or enhancing the ability of cells to generate energy in cardiometabolic disorders, where energy dysfunction is a root cause of disease pathogenesis, symptoms, and functional deficits. The lead candidate, ninerafaxstat, has completed multiple Phase 2 clinical trials across three indications: non-obstructive hypertrophic cardiomyopathy (nHCM), stable angina, and heart failure with preserved ejection fraction (HFpEF). In Phase 1 and Phase 2 clinical trials, ninerafaxstat demonstrated good tolerability. It represents an innovative treatment approach for cardiac conditions characterized by an imbalance between cardiac energy supply and demand. To maintain normal contractile function, the heart requires a significant amount of energy, primarily generated by mitochondria in the form of adenosine triphosphate (ATP). The heart typically uses two main fuels for energy production: fatty acids and glucose. Ninerafaxstat is a partial fatty acid oxidation (pFOX) inhibitor that shifts the heart’s preference from fatty acids to glucose. This metabolic shift results in more efficient mitochondrial energy production and has the potential to improve cardiac function at rest and during exercise. Ninerafaxstat is a simple oral compound that does not require dose titration or monitoring, has no clinically meaningful drug interactions, and can be used alongside standard cardiovascular therapies.
