Purple Biotech is a clinical-stage company dedicated to developing first-in-class therapies that overcome tumor immune evasion and resistance. The company's oncology pipeline includes NT219, CM24, and IM1240. NT219 is a dual inhibitor, a novel small molecule that simultaneously targets IRS1/2 and STAT3. In the phase 1/2 study of NT219, the company is currently conducting dose escalation as a monotherapy for solid tumors and combining it with cetuximab for the treatment of recurrent and metastatic squamous cell carcinoma of the head and neck (SCCHN) or colorectal cancer (CRC). Following these studies, an expansion cohort of NT219 will be conducted in patients with recurrent and metastatic SCCHN at the recommended phase 2 dose in combination with cetuximab. CM24 is a humanized monoclonal antibody that blocks CEACAM1, an immune checkpoint protein that supports tumor immune evasion and survival through multiple pathways. The company is advancing a combination therapy of CM24 with an anti-PD-1 checkpoint inhibitor into a phase 2 study for pancreatic ductal adenocarcinoma (PDAC). The company has entered into a clinical collaboration agreement with Bristol Myers Squibb for a phase 2 clinical trial to evaluate the combination of CM24 with the PD-1 inhibitor nivolumab and chemotherapy. IM1240 is a preclinical conditionally activated trispecific antibody that binds to T cells and NK cells, generating a potent localized immune response within the tumor microenvironment. IM1240 features cleavable capping technology that restricts the therapeutic activity of the compound to the local tumor microenvironment, potentially increasing the anticipated therapeutic window for patients.
