Roche Discontinues Phase 3 Development of Hemophilia A Gene Therapy SPK-8011, Advances Next-Gen FVIII Candidate

Roche recently announced that it will no longer continue the development of SPK-8011, a gene therapy for hemophilia A acquired during the 2019 acquisition of Spark Therapeutics.

According to the information from the clinical trial database ClinicalTrials.gov, the originally plannedSPK-8011 Gene Therapy DrugPhase 3 Clinical Trial KEYSTONE1 Marked as "Withdrawn" Due to Roche's Strategic Adjustment. Data Shows the Study Began Planning in March, but No Patients Were Enrolled.

Roche's spokesperson revealed to the media via email that the company has decided to halt further development of SPK-8011 and is preparing to launch a new, improved version of Factor VIII (FVIII) gene therapy as a new treatment option for Hemophilia A (reportedly, details of this drug have not yet been updated on the company’s official website pipeline). The spokesperson stated that this decision was made because they believe this improved version will be more effective.Coagulation Factor VIII (FVIII) Gene TherapyThe drug can better meet current medical needs while reducing the treatment burden on patients. Additionally, this decision is based on the favorable results of safety, durability, and predictability demonstrated by SPK-8011 in early studies, which primarily evaluated the safety and efficacy of low-dose gene therapy for patients with Hemophilia A.

SPK-8011 uses an adeno-associated virus (AAV) vector to deliver a normalCoagulation Factor VIII (FVIII)Gene delivery to the patient's liver cells. In this way, the patient’s liver cells can begin producing functional human FVIII protein, thereby helping restore normal blood clotting processes. The goal of this one-time treatment is to reduce or eliminate the need for regular clotting factor replacement therapy in the long term, improve the patient’s quality of life, and reduce the risk of disease-related complications.

Although the development of SPK-8011 has been terminated, its research has provided valuable data and experience for gene therapy in treating hemophilia A. Roche will continue to explore other innovative therapies with the aim of bringing more effective treatment options to patients with hemophilia.

Moreover, it is worth mentioning that Hemlibra, an antibody drug for hemophilia A from Roche.（Emicizumab) as early asIt was approved for marketing by the U.S. Food and Drug Administration (FDA) in 2017, and subsequently received regulatory approval in multiple countries and regions. Its introduction significantly improved the prognosis and quality of life for patients with Hemophilia A, becoming one of the key treatment options for patients.

Hemlibra is a bispecific antibody drug developed by Genentech, a subsidiary of Roche, for the treatment of Hemophilia A.

Hemlibra's uniqueness lies in its ability to simultaneously bind activated Factor IX and Factor X, simulating the function of Factor VIII, thereby helping restore the normal blood clotting process. This treatment is particularly important for Hemophilia A patients who have developed antibodies (inhibitors) against traditional replacement therapies, as their treatment options are extremely limited. Hemlibra, administered via subcutaneous injection, offers patients a more convenient and long-acting prophylactic treatment option.

Roche has high hopes for Hemlibra, viewing it as one of the key products in the company's hematology portfolio.
