On January 13, 2025, ZAIMING, an innovative oncology drug company under Simcere Pharmaceutical, announced that it had entered into a development and licensing option agreement with AbbVie for ZAIMING’s multiple myeloma (MM) candidate drug SIM0500.According to the terms of this agreement,ZAIMING will receive an upfront payment from AbbVie, as well as optional equity payments and milestone payments totaling up to $1.055 billion.ZAIMING will additionally receive tiered royalties based on the net sales of the product outside the Greater China region. AbbVie is entitled to receive tiered royalties on the net sales within the Greater China region.

SIM0500 isA humanized GPRC5D-BCMA-CD3 trispecific antibody developed by ZAIMING through its proprietary T-cell engager multispecific antibody technology platform.. This molecule combines a CD3 antibody with low affinity but high targeted activation, as well as two tumor-associated antibodies: anti-G protein-coupled receptor class C group 5 member D (GPRC5D) and anti-B cell maturation antigen (BCMA). SIM0500 exhibits potent T-cell cytotoxic effects against MM cells through multiple anti-tumor mechanisms. Currently,SIM0500Currently in Phase 1 clinical trials in both China and the United States.

In April 2024, the U.S. FDA granted SIM0500 Fast Track designation forPatients with multiple myeloma who have previously received ≥3 lines of therapy (including proteasome inhibitors, immunomodulatory agents, and anti-CD38 monoclonal antibodies), and are resistant to standard treatments known to provide clinical benefit, or cannot tolerate them.。

Multiple Myeloma is the second most common hematologic malignancy, capable of affecting multiple organs and tissues. In recent years, with the continuous advancement in diagnostic techniques and therapeutic drugs, significant progress has been made in the treatment of multiple myeloma over the past few years. However, many patients still face the challenge of relapse and refractory disease, and the difficulty of treating relapsed MM increases with the number of recurrences. These patients still need more effective, safer, and affordable treatment options.

Significant progress has been made in the use of immunotherapy for the treatment of relapsed/refractory multiple myeloma. Immunotherapy that directly targets surface antigens on tumor cells is a rapidly developing area in current MM treatment, with targets such as CD38, GPRC5D, and BCMA being highly expressed on malignant plasma cells, making them popular targets for MM immunotherapy.According to the introduction in the press release by ZAIMING,Tri-specific antibodies simultaneously target three antigens on tumor cells, more precisely exerting immune signal modulation, which may potentially be safer and more effective than combination therapies consisting of three mono-specific antibodies.
