On February 15, 2025, GSK announced that its pentavalent meningococcal vaccine Penmenvy received approval from the U.S. Food and Drug Administration (FDA) for use in individuals aged 10 to 25.This vaccine is able to prevent invasive meningococcal disease (IMD) caused by five common pathogenic strains of meningococcal bacteria: A, B, C, W, and Y.

Penmenvy combines the antigen components of GSK's two previous meningitis vaccines, Bexsero (for the prevention of meningococcal type B) and Menveo (for the prevention of meningococcal types A, C, W, and Y), offering more comprehensive protection. This synthetic vaccine simplifies the vaccination process by reducing the number of injections, which helps increase vaccination rates among adolescents and young adults.
The approval of Penmenvy is based on two Phase III clinical trials (NCT04502693 and NCT04707391) involving more than 4,800 participants aged 10 to 25. The trial results showed that the vaccine had good safety and effective immune response, consistent with the safety profile of GSK's existing meningitis vaccines. Specifically, Penmenvy elicited a strong immune response against 110 different MenB strains, covering 95% of the prevalent strains in the United States.
Meningococcal meningitis, although relatively rare, has a fatality rate as high as one in six, and delayed treatment can lead to severe sequelae such as brain damage, limb amputation, hearing loss, and neurological issues. Teenagers and young adults are at high risk for this disease due to living environments (such as dormitories and group activities) and behavioral habits (such as kissing and sharing eating utensils), which facilitate the spread of meningococcal bacteria.

Despite Pfizer's Pentavalent Vaccine Penbraya Receiving FDA Approval in October 2023, GSK's Penmenvy is Expected to Expand Its Market Share Due to Its Leading Position in the MenB Vaccine Market and Broader Protection. According to Evaluate Vantage, Penmenvy's Global Sales Are Predicted to Reach $1.1 Billion by 2030, While Penbraya is Forecasted at $606 Million.
GSK's Head of Medical Affairs, Cynthia Burman, stated that incorporating the MenB vaccine into comprehensive recommendations and simplifying the vaccination process with a pentavalent vaccine may help increase vaccination rates among adolescents and young adults. The Advisory Committee on Immunization Practices (ACIP) of the U.S. Centers for Disease Control and Prevention (CDC) plans to discuss GSK's Penmenvy vaccine at a meeting in late February 2025, potentially including it in the recommended U.S. immunization schedule.
