AstraZeneca announced today that the Phase 3 clinical trial MATTERHORN has achieved positive topline results.Its重磅PD-L1 inhibitor Imfinzi (durvalumab), in combination with the standard treatment FLOT (fluorouracil, leucovorin, oxaliplatin, and docetaxel) as a perioperative treatment regimen, demonstrated a statistically significant and clinically meaningful improvement in the primary endpoint of event-free survival (EFS).Patients received neoadjuvant treatment with Imfinzi combined with chemotherapy before surgery, followed by adjuvant treatment with Imfinzi combined with chemotherapy after surgery, and finally monotherapy with Imfinzi. This trial evaluated the efficacy of this regimen compared to perioperative chemotherapy alone in patients with resectable, early-stage, and locally advanced (Stage II, Stage III, Stage IVA) gastric cancer and gastroesophageal junction (GEJ) cancer.

In the interim analysis of the critical secondary endpoint pathological complete response rate (pCR) reported previously,The pCR rate of the Imfinzi combination treatment regimen was more than double that of neoadjuvant chemotherapy alone (19% vs. 7%, p<0.00001).
For the secondary endpoint of overall survival (OS), a clear trend favoring the Imfinzi regimen was observed in this interim analysis. The trial will continue to follow up with patients, and OS will be formally assessed in the final analysis. Detailed data will be presented at an upcoming medical conference and shared with global regulatory authorities.
Imfinzi is a humanized monoclonal antibody. By binding to PD-L1, it blocks the interaction between PD-L1 and PD-1 as well as CD80 proteins, thereby inhibiting the tumor's immune escape mechanism.
