Recently, YOUJIA BIO and SunNovo officially signed a project cooperation agreement and a strategic cooperation agreement. Together, they are committed to the research and development of an innovative Antibody Oligonucleotide Conjugate (AOC) new drug. Focusing on nucleic acid conjugate drug technology, they will concentrate on fields such as metabolic diseases and jointly advance the development of innovative drugs.
It is reported that the core of this cooperation between the two parties lies in the joint development of an innovative AOC drug. By utilizing the AOC technology platform, antibodies and siRNA drugs are ingeniously combined to achieve precise regulation of target molecules, with the aim of achieving better therapeutic effects. The two parties will make full use of their respective technical advantages and resources to deeply cooperate in various aspects such as drug discovery, research and development, clinical trials, and commercial promotion, jointly ensuring the smooth progress of the project.
It is reported that AOC drugs are gradually gaining popularity in the current pharmaceutical industry. In addition to the aforementioned companies, many other enterprises are also increasing their investments in this field through cooperation, financing, and other means.
On October 10, 2024, HitGen announced a strategic cooperation agreement with Chengdu Shengshi Junlian Biotechnology Co., Ltd. to integrate their trillion-level DNA Encoded Library (DEL), a complete small nucleic acid drug discovery and production platform, and a 300-billion-level biopharmaceutical molecule library, jointly providing Antibody-Oligonucleotide Conjugate (AOC) drug research and development services to meet the growing global demand for AOC drug R&D.
HitGen and AbLinker have previously collaborated on antibody, siRNA discovery optimization and preparation, Linker development, and AOC production, achieving certain results. Building on this, the two parties have decided to jointly offer AOC drug research and development technical services and AOC product licensing and transfer businesses.
Currently, although pharmaceutical companies in China are increasingly entering the field, globally, the development of AOC remains at a relatively early stage. It is mainly dominated by small and medium-sized biotech companies such as Avidity, Tallac Therapeutics, Dyne Therapeutics, Denali Therapeutics, and Gennao Bio in terms of early-stage research and development. The AOC projects developed by these companies target indications covering muscular diseases, central nervous system disorders, and cancer.
Among them, the first to enter clinical trials was Avidity's AOC drug. In November 2021, Avidity Biosciences' Antibody Oligonucleotide Conjugate (AOC) AOC 1001 entered Phase I clinical trials. This drug is a therapy for rare muscle diseases, utilizing the targeting ability of antibodies to deliver siRNA to muscle cells.
In addition, on May 20, 2024, Dyne Therapeutics announced new clinical progress for two oligonucleotide-conjugated antibody drugs (AOC). The data showed that both drugs significantly impacted disease biomarkers, improved function, and demonstrated good safety. Notably, Dyne's first AOC drug, DYNE-101, is currently undergoing the Phase I/II ACHIEVE trial in patients with type 1 myotonic dystrophy (DM1).
In the future, with the advancement of pharmaceutical companies' research and development, the clinical and commercial value of AOC will continue to become more prominent. Companies with mature platform technology, strong clinical development capabilities, and differentiated competitive advantages are expected to gain more market opportunities.
