On April 8, 2025, Eli Lilly registered a Phase I clinical trial on Clinicaltrials.gov for the combination therapy of amylin analog Eloralintide and tirzepatide in treating obesity or overweight.

This Phase I clinical trial plans to enroll 188 subjects and is expected to be completed by the end of the year.

Amylin is becoming a key player in the weight loss field, with Novo Nordisk advancing.Amylin in combination with semaglutide has obtained topline data from Phase III clinical trials for diabetes and weight loss. Amycretin, a GLP-1/Amylin dual-target drug, has also shown highly competitive efficacy in early clinical trials. The 1.25mg, 5mg, and 20mg dose groups achieved weight loss of 9.7%, 16.2%, and 22.0% respectively after 36 weeks of treatment, while the placebo group had a weight loss of approximately 2.0%.
On March 3, 2025, AbbVie announced the introductionDenmarkBiotechnology CompanyGubraGUB014295, an Amylin analog, under the agreement, AbbVie pays $350 million upfront, $1.875 billion in milestone payments, and a certain percentage of sales royalties.
On March 12, 2025, Roche announced a collaboration with Zealand to jointly develop the amylin analog Petrelintide.Roche paid $1.65 billion as an upfront payment, $1.2 billion in development milestone payments, and $2.4 billion in sales milestone payments, with the total agreement amounting to $5.3 billion.
Summary
Amylin has a different weight loss mechanism from GLP-1, tending to preserve muscle mass with better tolerability. It increases satiety rather than suppressing appetite, forming a perfect complement to single-target or multi-target GLP-1 drugs in terms of mechanism. Amid fierce competition among GLP-1 products, Amylin analogs, which have higher technical barriers, have become the focus of competition among MNCs.
