GSK's Single-Dose RSV Vaccine Demonstrates Sustained Protection Over Three Seasons in Older Adults

A latest study shows that GSK's RSV vaccine Arexvy (RSVPreF3 OA) provides effective protection for three consecutive RSV seasons with just a single dose in adults aged 60 and above.The research findings were published in *The Lancet Respiratory Medicine*.

This multicenter, randomized, observer-blind, placebo-controlled Phase III clinical trial (AReSVi-006) was conducted across 275 research centers in 17 countries worldwide, enrolling a total of 24,972 adults aged 60 years and above.

Participants were randomly assigned to receive one dose of Arexvy or a placebo before the first RSV season.Before the second season, participants who had been vaccinated were randomly assigned to receive a second dose of the vaccine or a placebo.

The research results show that the cumulative effectiveness of a single dose of the vaccine over three seasons was 62.9%, providing protection against both RSV A and RSV B subtypes.

In addition, the vaccine showed good protective effects across all age groups and in elderly people with underlying health conditions.The study also found that a repeat vaccination one year later did not significantly increase the overall effectiveness of the vaccine.

RSV is a highly contagious virus that primarily affects the lungs and respiratory tract, posing a serious threat to the elderly and people with chronic diseases.Each year, RSV causes approximately 60,000 to 120,000 hospitalizations among adults aged 65 and older in the United States, and 6,000 to 10,000 deaths.Arexvy vaccine was approved by the U.S. FDA in 2023 for the prevention of RSV-related lower respiratory tract diseases in individuals aged 60 years and above.

This study provides important evidence for the development of RSV vaccine vaccination strategies, indicating that a single dose may offer sustained protection across multiple seasons, reducing the disease burden in the elderly population.
