PF-08052666 adopts in its molecular designNovel antibodies, differentiated linker-payloads, and higher drug-to-antibody ratio (DAR), aiming to address the limitations of previous MSLN-targeted ADC therapies. This ADC consists of a fully human IgG1 monoclonal antibody linked to a potent TOP1i payload based on camptothecin through a protease-cleavable linker, with a DAR value averaging 8.

The main preclinical study results published this time include:

  • In vitro studies have shown that PF-08052666 directly exerts cytotoxic effects by delivering payloads to MSLN-positive cells and demonstrates killing activity against co-cultured MSLN-negative cells through a bystander effect.Even at physiological concentrations, soluble MSLNExistenceTheUnder the circumstancesPF-08052666 still possesses cell-killing capability.

  • In vivo study data show that PF-08052666 outperformed the DM4-based anti-MSLN benchmark ADC across various tumor cell lines and patient-derived xenograft models, including ovarian cancer, lung cancer, and colorectal cancer.

  • In a heterologous xenograft model composed of a mixture of MSLN-positive and MSLN-negative cells, PF-08052666 also demonstrated superior efficacy compared to the DM4-based anti-MSLN benchmark ADC, indicating that the novel linker-payload of MesoC2 has a more significant bystander effect.


The highly promising preclinical study results provided strong support for the initiation of the first-in-human Phase I clinical trial of PF-08052666 in patients with advanced solid tumors.Currently, the trial (NCT06466187) is undergoing participant recruitment.
