AstraZeneca recently announced positive results from two Phase III clinical trials (KALOS and LOGOS) for its triple-combination inhaler Breztri Aerosphere (budesonide/glycopyrronium bromide/formoterol fumarate), demonstrating significant efficacy in patients with uncontrolled asthma. This outcome is expected to expand the indications for Breztri Aerosphere from chronic obstructive pulmonary disease (COPD) to the treatment of asthma.

KALOS and LOGOS Trials Are Two Randomized, Double-Blind Phase III Clinical Studies, Enrolling Approximately 4,400 Adult and Adolescent Patients with Uncontrolled Asthma. The Results Showed That Compared to the Traditional Inhaled Corticosteroid/Long-Acting Beta-2 Agonist (ICS/LABA) Dual Therapy, Breztri Aerosphere Demonstrated Statistically Significant and Clinically Meaningful Improvements in Lung Function, Specifically Evidenced by a Significant Improvement in Forced Expiratory Volume in One Second (FEV1) from Baseline at Week 24.No new safety signals were identified in the trial.

Breztri Aerosphere has been approved in over 80 countries, including the United States, the European Union, China, and Japan, for the treatment of COPD. In 2024, the drug's sales increased by 44%, reaching $978 million. If its indications are expanded to include asthma treatment, it is expected to further boost its market performance, with potential peak revenues ranging from $3 billion to $5 billion.

AstraZeneca plans to submit the full results of these two Phase III trials to regulatory authorities and release them at an upcoming medical conference.

Asthma is a common chronic respiratory disease that affects approximately 262 million people worldwide. Among them, nearly half of the patients receiving dual therapy still lack effective control, leading to limited lung function and a reduced quality of life. The breakthrough progress of Breztri Aerosphere has brought new hope to these patients.

Conclusion:

Breztri, along with the IL-5 inhibitor benralizumab and the TSLP inhibitor tezepelumab, forms AstraZeneca's core portfolio for growth in the respiratory disease field. The latter two drugs are both approved for severe asthma indications and are also under development for COPD. Last year, AstraZeneca reported new data on Fasenra for acute asthma, calling it the first major advancement in this aspect of the disease in 50 years.

AstraZeneca's ambitious goal is to increase group revenue from over US$54 billion in 2024 to more than US$80 billion by 2030, with Respiratory being a key component of this target. The company has high hopes for Breztri, expecting it to generate between US$3 billion and US$5 billion in revenue at its peak.
