Efimosfermin alfa is a long-acting FGF21 analog administered via monthly subcutaneous injection, with multiple potentials including metabolic regulation, hepatic lipid reduction, inflammation improvement, and fibrosis reversal.Phase II clinical trial data shows, the drug can rapidly andSignificantly Reverse the Process of Liver Fibrosis in Patients with Moderate to Advanced MASH, the efficacy is independent of whether GLP-1 background therapy is used, demonstrating its competitiveness as a potential best-in-class treatment. Its good tolerability, low immunogenicity, and extended half-life also provide possibilities for monthly dosing and improved patient compliance.

In addition, efimosfermin alfa can reduce triglycerides and improve glycemic control, potentially offering additional benefits for MASH patients with cardiometabolic syndrome.Its mechanism complements GSK's self-developed siRNA therapy GSK4532990, and in the future, combination therapy could potentially cover a broader subpopulation of patients with fatty liver disease.

Steatotic Liver Disease (SLD) affects approximately 5% of the global population, representing a field with limited treatment options and a significant healthcare burden. Among these, MASH and alcoholic liver disease (ALD) have become the leading causes of liver transplantation in the United States. Effectively halting the progression of mid- to late-stage fibrosis could save the U.S. healthcare system between $40 billion and $100 billion over the next 20 years.
