Lilly's GLP-1 Drug Orforglipron Falls Short of Semaglutide in Phase III Obesity Trial, Wiping Out $100 Billion in Market Cap

On August 7, 2025, Eli Lilly and Company released its second-quarter financial report and updated the topline data from the Phase III clinical trial ATTAIN-1 for the small molecule GLP-1 receptor agonist Orforglipron in treating obesity. The 36mg high-dose group achieved an 11.5% weight loss after 72 weeks of treatment (placebo-adjusted).


In contrast, the weight loss achieved with the 2.4mg dose of semaglutide after 72 weeks of treatment was 15.1% (placebo-corrected).


Affected by this news, Eli Lilly's stock price plummeted 14% in a single day, with its market value dropping from $700 billion to $600 billion. Orforglipron alone triggered a loss of $100 billion in market value.


Peter Cardillo, Chief Market Economist at Spartan Capital Securities, stated that the Eli Lilly incident reflects the market's sensitivity to earnings and R&D progress in a high-valuation environment. "The boost from the earnings season remains, but negative news on individual stocks can easily trigger a rapid adjustment in the sector."
