On August 1, 2025, Amgen registered a Phase I clinical trial for AMG 410 on the Clinicaltrials.gov website.

This Phase I clinical trial plans to enroll 434 patients with advanced solid tumors carrying KRAS mutations, with preliminary completion expected by September 2027.

AMG 410 is a non-covalent pan-KRAS inhibitor that simultaneously binds to GDP (off) and GTP (on). This molecule binds to the same allosteric pocket as KRAS G12C inhibitors but exhibits high potency in KRAS G12D, G12V, G12C, G13D, and other mutants. AMG 410 shows higher selectivity for KRAS over HRAS and NRAS, avoiding cytotoxicity to non-KRAS-transformed cells.
