This trend indicates that RNAi technology is becoming a crucial direction in the research and development of innovative drugs worldwide, with cross-border cooperation being key to accelerating its commercialization process.

Argo Biopharma Deepens Collaboration with Novartis to Advance Cardiovascular siRNA Therapy
On September 3, Argo Biopharma announced a new strategic collaboration agreement with Novartis to jointly develop multiple cardiovascular siRNA therapies. Under the agreement, Argo Biopharma will receive an upfront payment of $160 million and is eligible for up to $5.2 billion in milestone payments and sales royalties.
Argo Grants Novartis Option for Rights Outside China to Two Early-Stage Molecules for the Treatment of Severe Hypertriglyceridemia (sHTG) and Mixed Dyslipidemia, as well as Priority Negotiation Rights for BW-00112 (ANGPTL3). The product is currently in Phase 2 clinical trials in the United States and China, with subsequent combination therapy clinical trials to be led by Argo.For another siRNA candidate drug in the preclinical research stage, Argo has granted Novartis an exclusive license outside of China, with a reciprocal option to share profits and losses (P&L) in the United States and China. This product is expected to initiate Phase 1 clinical trials in 2026.
Arrowhead Collaborates with Novartis to Develop siRNA Therapies for Neurodegenerative Diseases
On September 2, Arrowhead Pharmaceuticals announced a global licensing and collaboration agreement with Novartis, granting Novartis exclusive rights to Arrowhead's siRNA therapy ARO-SNCA for the treatment of neurodegenerative diseases such as Parkinson's disease. Under the agreement, Arrowhead will receive an upfront payment of $200 million and is eligible for up to $2 billion in milestone payments and sales royalties.
ARO-SNCA is an siRNA therapy targeting the α-synuclein gene, designed to inhibit the abnormal aggregation of pathogenic proteins through the RNAi mechanism. Arrowhead's TRiM platform has shown excellent performance in preclinical studies, achieving delivery to the central nervous system, including deep brain regions, via subcutaneous administration. This technological breakthrough offers new possibilities for the treatment of neurodegenerative diseases, particularly for gene targets that are difficult to address with traditional drugs.
AbbVie &ADARxReach siRNA Therapy Development Cooperation
On May 14, AbbVie and ADARx Pharmaceuticals announced a collaboration and license option agreement aimed at jointly developing siRNA therapies for multiple disease areas, including neuroscience, immunology, and oncology.According to the terms of the agreement, ADARx will receive an upfront payment of $335 million and is eligible for billions of dollars in milestone payments.
This strategic collaboration will fully leverage ADARx's expertise in RNA discovery and its proprietary siRNA technology, which has the potential to achieve durable and precise mRNA silencing. AbbVie, on the other hand, will provide professional support in areas such as antibody engineering, antibody-drug conjugates, and tissue delivery strategies to assist ADARx in its drug discovery efforts.
Biogen Reaches Over $1 Billion RNAi Therapy Collaboration Agreement
On May 27, Biogen and City Therapeutics reached a strategic cooperation agreement to jointly develop several novel RNAi therapies.According to the terms of the agreement, City Therapeutics will receive a total of $46 million, including a $16 million upfront payment and up to $1 billion in milestone payments.
According to the agreement, City Therapeutics will utilize its next-generation RNAi engineering technology to develop RNAi-triggering molecules, which will be combined with Biogen's proprietary drug delivery technology. The initial focus of this collaboration will be on a key target that plays an important role in central nervous system diseases. The project will adopt tissue-enhanced delivery technology with the goal of achieving systemic administration. Under the terms of the agreement, Biogen will be responsible for all preclinical studies required for IND, global clinical development, regulatory submissions, and subsequent commercialization-related activities.
Jingyin Pharmaceuticals on Long-Acting siRNA New DrugWith CRISPR TherapeuticsReach Authorization Cooperation
On May 20, Argo Biopharma and CRISPR Therapeutics established a strategic partnership to jointly advance the co-development and commercialization of siRNA therapies.The two parties will jointly develop SRSD107. In addition, the agreement grants CRISPR Therapeutics the right of first negotiation for the exclusive licensing of up to two additional siRNA projects in the future.According to the terms of the agreement, Jingyin Pharmaceuticals will receive $95 million in cash and cash equivalents as an upfront payment from CRISPR Therapeutics and is eligible to receive over $800 million in advance and milestone payments.
This collaboration combines the complementary capabilities of both parties in R&D and industrialization to jointly develop and commercialize SRSD107 for the treatment of thrombotic and thromboembolic diseases.SRSD107 Injection is a double-stranded small interfering nucleic acid (siRNA) drug designed to selectively inhibit coagulation factor XI (FXI). The potential indications for SRSD107 are broad, including atrial fibrillation, venous thromboembolism (VTE), cancer-associated thrombosis, patients with end-stage renal disease undergoing hemodialysis, and individuals undergoing major orthopedic surgery whose treatment options are limited by bleeding risks.Currently, SRSD107 has completed two Phase 1 clinical trials.
