Enitiatebio, a Hangzhou-based biopharmaceutical company, has secured a significant regulatory milestone for its dual-target drug candidate QY201, which has been included in China's Breakthrough Therapy Designation program for treating adults with moderate-to-severe atopic dermatitis.

The announcement, made on October 23, marks a critical step forward for the company's lead asset—a next-generation highly selective dual JAK1/TYK2 inhibitor designed to block the JAK-STAT inflammatory signaling pathway and downregulate multiple key pro-inflammatory cytokines.

Atopic dermatitis, a chronic inflammatory skin condition affecting millions worldwide, has limited treatment options for patients with moderate-to-severe disease. The condition causes intense itching, skin lesions, and significant impairment to quality of life, creating substantial unmet medical need.

In a completed Phase 2 clinical trial for moderate-to-severe atopic dermatitis, QY201 demonstrated significant efficacy across multiple key endpoints, including EASI-75, EASI-90, and IGA response rates. The drug showed rapid and sustained itch relief, efficient clearance of skin lesions, and meaningful improvements in patients' clinical symptoms, signs, and overall quality of life. Safety and tolerability profiles were favorable, the company reported.

The program has now advanced to Phase 3 pivotal registration trials. The ongoing multi-center, randomized, double-blind, placebo-controlled study has enrolled 461 subjects. Primary endpoints include EASI-75 response rate and IGA response rate at Week 16, data that will be critical for regulatory approval.

China's Breakthrough Therapy Designation program, administered by the Center for Drug Evaluation under the National Medical Products Administration, provides expedited review pathways for innovative drugs addressing serious conditions with significant clinical advantages. The designation typically accelerates development and review timelines, potentially bringing promising therapies to patients more quickly.

The dual JAK1/TYK2 inhibition mechanism represents a novel approach in the atopic dermatitis treatment landscape. By targeting two key inflammatory pathways simultaneously, QY201 aims to provide more comprehensive disease control compared to single-target therapies currently available in the market.

Enitiatebio's progress comes at a time when the atopic dermatitis market is experiencing significant growth, driven by increasing prevalence, greater disease awareness, and the emergence of targeted therapies. The success of QY201 in Phase 3 trials could position Enitiatebio as a meaningful player in this competitive therapeutic area.
