A KRAS-EGFR Dual-Targeting ATTC Drug Goes Global! HUTCHMED and GSK Reach a Worldwide License Agreement

On September 3, HUTCHMED announced that its subsidiary HUTCHMED Limited had entered into an exclusive license agreement with a subsidiary of GSK plc, granting the GSK subsidiary the rights to develop and commercialize HMPL-A830 worldwide, excluding Mainland China, Hong Kong, Macau, and Taiwan.

HMPL-A830 is a first-in-class antibody-targeted conjugate (ATTC), formed by conjugating a highly selective, highly potent KRAS small-molecule inhibitor payload to an anti-EGFR antibody. Through antibody-guided delivery, the design selectively transports the potent payload to EGFR-expressing tumor tissue while simultaneously blocking both the EGFR and KRAS signaling pathways, with the aim of improving efficacy, durability, and tolerability. Colorectal, pancreatic, and lung cancers are the tumor types with the highest prevalence of KRAS gene alterations, and existing therapies often lack safe, durable solutions — the unmet need HMPL-A830 targets.

Under the agreement, HUTCHMED Limited will receive an upfront payment of USD 110 million and is eligible for up to USD 1.185 billion in development, regulatory, and commercial milestone payments, plus tiered royalties based on net sales. HUTCHMED retains all R&D and commercialization rights to the drug in Greater China. In addition, GSK holds priority negotiation rights for another, earlier-stage ATTC candidate from HUTCHMED. The transaction is subject to customary closing conditions, including antitrust review.

Clinical development will initially focus on colorectal, pancreatic, and lung cancers. HUTCHMED Limited will lead the global Phase I study of HMPL-A830, which is expected to start in H2 2026 (registration number: NCT07718581); GSK will lead all subsequent clinical development and commercialization outside Greater China.

Zheng Zefeng, interim chief executive officer and chief financial officer of HUTCHMED, said this collaboration is a key step in maximizing the drug's clinical potential and will open a new chapter in precision oncology. Dr. Hesham Abdullah, senior vice president of GSK and head of global oncology R&D, noted that HMPL-A830's dual KRAS-EGFR targeting mechanism has the potential to significantly improve upon current standard-of-care regimens.
