EG-501 is a small molecule oral tablet for the treatment of cognitive impairment in neuropsychiatric systemic lupus erythematosus (NPSLE). Cognitive impairment, commonly known as "brain fog," presents as a series of symptoms including lack of focus, memory decline, and slow thinking. As one of the common complications of lupus, cognitive impairment severely affects patients' quality of life and social functioning, yet this area...No approved drugs are currently available., there is a huge unmet clinical need. EG-501 has undergone a multi-center, randomized, placebo-controlled, double-blind Phase II trial at three renowned academic centers including Vanderbilt University Medical Center in the United States. The trial aims to evaluate its efficacy and safety in patients with NPSLE cognitive impairment and has achieved breakthrough positive results.
As one of the core pipelines deeply involved by Evergreen Therapeutics' self-developed AI platformThe EG-501 project started with AI modeling and innovatively applied machine learning algorithms such as PheWAS analysis, LASSO, and Random Forest to deeply explore the associations between targets and diseases. The project introduced large-scale high-quality data from top-tier academic medical centers in the United States, including whole-genome data covering more than 12,000 exome sequencing results, 250,000 genetic coding variants, and target-related SNPs. The de-identified medical case database contains information on 35,000 subjects, covering approximately 1,600 different disease phenotypes. Based on this, the platform used precise feature selection to identify 12 disease phenotypes related to the target from massive amounts of data, further combining market demand with commercial value assessment.Finally, it was determined to use NPSLE cognitive impairment as the indication for Phase II clinical trials, establishing a complete technical closed-loop from data to clinical direction.In the design of clinical trial protocols, the application of AI technology has significantly improved the efficiency of selecting enrolled patients. By precisely targeting the desired patient population, it achieves statistical significance with fewer enrolled patients, effectively reducing trial costs.
