On October 21, focused onNewLimit, a company focused on epigenetic reprogramming, secures additional $45 million in funding, with investors including Eli Lilly, Duke University Management Company, Section 32, Abstract, etc. This round of financing is due to the technical breakthrough in their project to restore youthful liver function, with plans to initiate clinical research in the coming years.

NewLimit was founded in 2021 by Brian Armstrong, co-founder of the cryptocurrency platform Coinbase, and former GV.(Google Ventures)Founded by partner Blake Byers. In May this year, the company had just completed a $130 million Series B financing round.

NewLimit's core proposition is: The decline in cellular function caused by aging is the root cause of various diseases and a decrease in quality of life. If the function of aging cells can be reversed, these age-related issues can be specifically addressed. The aging process, once considered irreversible, has been shown to be malleable through epigenetic reprogramming technology, based on the 2012 Nobel Prize-winning discovery —Shinya Yamanaka factors can revert mature cells to youthful pluripotent cells

However, the factors discovered by Shinya Yamanaka at the time posed a risk of inducing tumors. NewLimit has since focused on screening safer epigenetic reprogramming factors for different cell types and indications. The company’s R&D process begins with constructing tools to detect cellular functional aging, followed by leveraging AI reprogramming models, genomic screening, and functional validation to identify therapeutic transcription factor combinations, ultimately developing RNA-based drugs.

It is reported that NewLimit's core pipeline isUsing lipid nanoparticles(LNP)Delivery of mRNA to Hepatocytes, the transcription factor encoded by this mRNA is known to make hepatocytes appear and function younger. This candidate product is currently in the preclinical research and development stage. However, its ability to secure funding based on its technical platform and preclinical candidate products reflects the capital's recognition of its prospects. Its accumulation of LNP delivery technology in the field of hepatocytes can be extended to various cell types such as immune cells and endothelial cells.

Anti-aging Financing

Anti-aging has been a hotly debated topic in recent years. Under extremely high attention, the "anti-aging × innovative pharmaceuticals" field has also developed rapidly. Since the beginning of this year, in addition to NewLimit, several anti-aging innovative pharmaceutical companies have received financing.

In July this year, Foundry BioSciences, which focuses on generative AI-enabled protein design for anti-aging products and drug development, officially completed its Pre-Seed round of financing.Total financing reached tens of millions of RMB. The funds will be mainly used for the continuous upgrade of the technology platform, the expansion of the core team, and the advancement of industrial transformation for multiple ongoing research projects, further promoting the cutting-edge application of AI-driven protein language models in longevity science.

In May this year, Juvenescence announced the first closing of its B-1 round led by cornerstone investor M42.Has raised $76 million in funding, and received follow-on investments from existing investors. The company focuses on anti-aging research and the development of therapies for age-related diseases, exploring diverse technological pathways including small molecule drugs, gene therapy, and regenerative medicine.

Retro Biosciences Focuses on the Development of Cell Reprogramming and Autophagy Technologies,Announced the launch of a $1 billion Series A round of financing earlier this year, while OpenAI CEO Altman invested $180 million in the company in his personal capacity. Notably, the co-founders of Retro Biosciences include Ding Sheng, former dean of the School of Pharmaceutical Sciences at Tsinghua University and a scholar in the field of stem cell research.

In August this year, Retro and OpenAI announced that they had successfully used GPT-4b micro to design protein variants of the Yamanaka factors. Can increase reprogramming efficiency by 50 timesIn the study, the modified Yamanaka factors successfully yielded healthy, fully reprogrammed stem cells.

In the same month, Ningmei Haowei Biotech announced the completion of an angel round of financing worth tens of millions. The funds will be used for Ningmei Haowei.Research and Industrialization Progress in the Field of Gene-Edited Cell Therapy, supporting its clinical application in the fields of precision anti-aging and chronic disease treatment such as diabetes.

In addition, Klotho, a star company in longevity gene therapy, announced this year that it would expand its R&D pipeline from the field of neurodegenerative diseases to broader anti-aging areas such as muscle strength and bone health. It plans to build an all-around "longevity platform" by acquiring complementary technologies, delivering secreted Klotho protein via AAV vectors.(s-KL)Successfully Extended Mice Lifespan by 20%, and significantly improved the functional indicators of the heart, liver, kidneys, and brain. This also caused the company's stock price to soar 787% in a single day, with trading volume exceeding 1.1 billion shares.

Diversified Anti-Aging Methods

In the field of innovative biopharmaceuticals, there are various anti-aging methods, including immunocyte therapy, stem cell therapy, and gene therapy.

In immunocyte therapy,Current research mainly focuses on CAR-T targeting uPAR.For example, the research paper "Prophylactic and long-lasting efficacy of senolytic CAR T cells against age-related metabolic dysfunction" reported a uPAR-targeted CAR-T that can safely and selectively eliminate senescent cells, improve motor function in aged mice, and ameliorate metabolic dysfunction in both aged mice and high-fat diet mice, rejuvenating aged mice or slowing down aging in young mice. In China, Huanma Biotech has also conducted related research on this target for CAR-T.

In the field of stem cells,This therapy mainly focuses on the direction of repair and regeneration.Stem cells, with their powerful self-renewal and multi-directional differentiation potential, can differentiate into various tissue cells, replenish aging or damaged cells, activate the body's self-healing mechanism, and fundamentally assist in the repair and regeneration process of the body, delaying aging. For example,Hamilton BioHuman umbilical cord mesenchymal stem cell injection has been approved by the CDE to enter clinical trials, with the indication being premature ovarian failure, becoming the first domestically produced stem cell candidate drug in China to be approved for clinical trials for this indication.

In the field of gene therapy,Core Focus: "Regulation of Aging Mechanisms" and "Reversal of Senescent Cell Function"By precisely intervening in gene expression or repairing aging-related gene defects, the aging process can be delayed or even reversed at the molecular level. It is mainly divided into three major technical directions: epigenetic reprogramming gene therapy, aging-related gene editing therapy, and longevity gene delivery therapy. Klotho falls under the third category.
