【Pharmaceutical Network Industry DynamicsCDH17, as a transmembrane adhesion molecule, is highly restricted to the basolateral membrane expression in normal intestinal epithelial tissues, while showing significant overexpression in digestive tract malignancies such as colorectal cancer, gastric cancer, and pancreatic cancer. This unique expression pattern is quickly making CDH17 a rising star in the field of gastrointestinal tumor treatment. Recent developments in this area have been frequent.

In October, Mabwell's innovative ADC drug 7MW4911 targeting CDH17 initiated a Phase I/II clinical study to evaluate its safety, tolerability, pharmacokinetic characteristics, and preliminary efficacy in patients with advanced solid tumors. Advanced solid tumors refer to malignant growths in body organs or tissues that have progressed to an advanced stage, with symptoms varying depending on the tumor location, such as pain, lumps, etc. The primary objectives of this trial are to assess the safety and tolerability of 7MW4911 in patients with advanced solid tumors, determine the MTD, RED, and RP2D, and preliminarily evaluate its efficacy and OBD in selected solid tumor patients. Secondary objectives include evaluating its pharmacokinetic profile, preliminary efficacy, and immunogenicity characteristics.

In October, Hansoh Pharma announced that it would license the global rights outside of Greater China for its investigational CDH17-targeted antibody-drug conjugate (ADC) HS-20110 to Roche. According to available information, HS-20110 is an investigational CDH17-targeted antibody-drug conjugate (ADC) currently in global Phase I clinical trials in China and the United States for the treatment of colorectal cancer (CRC) and other solid tumors.

According to the licensing agreement, the licensor will grant the licensee an exclusive global license to develop, manufacture, and commercialize HS-20110 ("the product"). The licensor will receive an upfront payment of $80 million and is eligible to receive up to $1.45 billion in milestone payments based on the development, regulatory approval, and commercialization progress of the product, as well as tiered royalties on future potential product sales.

These two events are a microcosm of Chinese pharmaceutical companies' efforts in the CDH17 field. According to statistics, there are currently at least 9 CDH17-targeted ADC drugs that have entered clinical trials or the IND application stage globally, all of which originate from Chinese pharmaceutical companies, fully demonstrating China's R&D strength in this cutting-edge target area.

From the perspective of market value, the CDH17 track holds enormous potential. Data shows that in 2023, the global colorectal cancer drug market size was approximately $13.8 billion, while the market size for colorectal cancer drugs in China was about 29.7 billion RMB. Moreover, the overseas licensing model allows Chinese pharmaceutical companies to share in the global market红利 in advance, with the collaboration between Hansoh Pharma and Roche serving as a typical example.

Currently, the CDH17 track is in a critical phase of clinical validation. Whether it can continuously verify safety and efficacy in large-scale trials will determine if it becomes another重磅 target. For Chinese pharmaceutical companies, it is necessary to accelerate clinical progress while also emphasizing basic research to address the challenge that the carcinogenic mechanism of CDH17 has not been fully elucidated.

The recent flurry of activity in the CDH17 field has witnessed the innovative transformation of China's pharmaceutical industry. Looking ahead, as more innovative drugs enter clinical trials, this target will not only bring new hope to patients with gastrointestinal tumors but also propel Chinese pharmaceutical companies to a central role in the global precision medicine arena.
