First In Vivo Gene Editing Therapy Releases Initial Data
On September 6, US biotechnology company Sangamo announced early data from its in vivo gene-editing therapy SB-913 for Hunter syndrome (also known as MPS II). Two patients receiving low doses showed no response, but two patients receiving medium doses exhibited significant reductions in glycosaminoglycan (GAG) levels after 16 weeks of treatment with SB-913 (urinary GAGs decreased by 51%, dermatan sulfate by 32%, and heparan sulfate by 61%). However, the enzyme expressed by the gene (IDS) was not detected in peripheral blood. Sangamo stated that the reduction in GAGs is sufficient to indicate repair of the IDS gene; the non-detection of IDS may be attributed to insufficient sensitivity of current detection methods or to the possibility that IDS was synthesized but rapidly taken up by target tissues.
