ASLAN Pharmaceuticals Secures Global Rights to ASLAN004 for Atopic Dermatitis and Other Indications

VCBeat (WeChat Official Account: vcbeat) learned from foreign media that biopharmaceutical company ASLAN Pharmaceuticals recently announced it has entered into a licensing agreement with biotechnology company CSL Limited (CSL) for the fully human antibody ASLAN004. Under the agreement, ASLAN Pharmaceuticals retains the rights to develop, manufacture, and commercialize ASLAN004 for all indications globally. Reportedly, this new agreement replaces the previous licensing agreement signed by both parties in May 2014.

 

Under the terms of this agreement, ASLAN will make an initial payment of $30 million to CSL upon the commencement of Phase 3 clinical trials for ASLAN004. CSL is also eligible to receive up to $95 million in regulatory milestone payments and $655 million in sales milestone payments, as well as tiered royalties on net sales. Under the original agreement terms, ASLAN was responsible for the development of ASLAN004, while CSL was entitled to 40% to 50% of the total gross revenue from ASLAN004, including proceeds from out-licensing agreements.

 

ASLAN004 is a fully human monoclonal antibody that targets the interleukin-13 receptor alpha 1 subunit (IL-13Rα1). By targeting IL-13Rα1, ASLAN004 effectively inhibits the signaling pathways of interleukin-4 (IL-4) and interleukin-13 (IL-13), which are key drivers of the allergic symptoms associated with atopic dermatitis. ASLAN Pharmaceuticals is currently conducting Phase I clinical trials in healthy volunteers to evaluate ASLAN004 as a treatment for atopic dermatitis.

 

Atopic dermatitis is the most common skin disease, affecting over 200 million patients worldwide. It is characterized by red, swollen, and inflamed skin accompanied by severe itching, which significantly impairs patients' quality of life. Currently, up to one-third of adult patients with atopic dermatitis are diagnosed with moderate-to-severe disease. Available treatment options are limited, and providing regular care for these patients is often challenging.

 

Based on the recently released data from the ASLAN004 trial, intravenous administration of ASLAN004 was found to be safe and well-tolerated at all dose levels. Researchers analyzed downstream mediators in patients following ASLAN004 injection, specifically assessing the phosphorylation status of STAT6, a key mediator of allergic inflammation. They observed that patient symptoms were suppressed within one hour after dosing and sustained for more than 29 days.

It is reported that ASLAN Pharmaceuticals plans to initiate clinical trials in patients with moderate-to-severe atopic dermatitis in the second half of 2019, conducting multiple ascending-dose studies of ASLAN004.
