MHC Tetramer Staining technology is widely used for the detection and analysis of antigen-specific T cells. 1 Ultra-rapid preparation. Polaris Biology offers unique unloaded MHC tetramer products, enabling the preparation of various peptide-MHC tetramers in just 30 minutes with a peptide loading efficiency of over 90%;
2. Wide variety of MHC types and label options. The product catalog includes over 300 pre-made antigen-MHC tetramers, offering a choice of 19 fluorescent detection channels and 40 mass cytometry metal detection channels; tetramer staining experiments can be easily performed on flow cytometers and mass cytometers.

In fields such as immunology research, antibody drug development, and vaccine development, high-throughput analysis of peptide-MHC complex and T-cell receptor (TCR) pairing, as well as T-cell function, is often required. However, due to limitations in the preparation time and cost of peptide-MHC tetramers, classic tetramer products are difficult to prepare rapidly, often requiring dozens of hours to complete antigen loading. Furthermore, the limited number of detection channels in conventional flow cytometry restricts its broad application. Mass cytometry technology achieves a qualitative leap in channel capacity by quantifying mass tags via metal ion systems. Through barcode design, it enables simultaneous high-throughput screening of T-cell antigen epitopes and multiparametric “phenotype + function” analysis of T cells.
