On March 20, Novartis announced that it had reached an agreement with Synnovation Therapeutics to acquire its pan-mutant selective PI3Kα inhibitor SNV4818.
According to the terms of the agreement, Novartis will pay a $2 billion upfront payment and provide up to $1 billion in milestone payments to acquire Pikavation Therapeutics, a wholly-owned subsidiary of Synnovation Therapeutics, which holds a portfolio of pan-mutation selective PI3Kα inhibitor projects, including SNV4818. The transaction is expected to be completed in the first half of 2026.
SNV4818 is an orally administered drug currently in Phase I/II clinical trials for the treatment of breast cancer and other advanced solid tumors. The biological mechanism of mutated PI3Kα in HR+/HER2- breast cancer indicates that approximately 40% of patients with this subtype may face a poorer disease prognosis due to the presence of PIK3CA gene mutations in their tumors. SNV4818Can be used as part of a potential combination therapy regimen, synergistically with CDK inhibitors and endocrine (hormonal) therapies.
Current PI3Kα inhibitors block both mutant and wild-type enzymes, leading to tolerability challenges that make it difficult for patients to adhere to treatment. SNV4818 focuses on the mutant form in tumors, aiming to reduce side effects, support more consistent dosing, and facilitate easier combination with earlier treatment regimens. Preclinical studies show,SNV4818It has potent activity against common PIK3CA mutations and demonstrates clear selectivity for the normal enzyme.
