Small-molecule chemical drugs long dominated the pharmaceutical industry during the era of generic drugs. China’s recent wave of innovative drug development also began with a few follow-on innovative small-molecule targeted therapies.

Small-molecule innovative drugs remain the cornerstone of drug development. Driven by population aging, the burden of chronic non-communicable diseases, particularly malignant tumors, has intensified, giving rise to substantial unmet clinical needs and propelling China’s small-molecule innovative drug industry onto a fast track. In 2021, the global market size for small-molecule innovative drugs was approximately USD 180 billion, with China’s market reaching about USD 40 billion, accounting for more than 20% of the global market.

Numerous outstanding enterprises have emerged in China’s small-molecule innovative drug industry, including established large-scale generic drug manufacturers transitioning toward innovative drug R&D, innovative pharmaceutical companies that have achieved leapfrog development through early-stage technological accumulation, industrial upgrading, or business model innovation, and a multitude of clinical-stage biotech startups engaging in differentiated competition based on their diverse product portfolios and technical approaches. Together, these entities constitute the industry ecosystem for small-molecule innovative drugs in China.

On June 17, 2022, the 2022 Small-Molecule Innovative Drug Development Forum will be held online. The forum will focus on the latest hot topics in small-molecule innovative drugs, bringing together leading enterprises and investors to conduct an in-depth analysis of the prospects, directions, bottlenecks, and future challenges in the development of small-molecule drugs from multiple perspectives, including target selection, technological pathways, and investment outlooks. By examining exemplary cases in the biopharmaceutical sector, the forum aims to highlight the courage and wisdom of pioneers in new drug development, guide the industry in seizing the historic opportunities for pharmaceutical innovation in China over the next decade, and provide impetus for the growth of the small-molecule innovative drug industry.

Join the Subforum on Innovative Small-Molecule Drug Development to engage with industry leaders on three hot topics in small-molecule innovation, gain firsthand insights into cutting-edge achievements and expert opinions, and jointly explore the future of innovation in China!

How Can Chinese Innovative Small-Molecule Drugs Embark on a New Journey?

China’s new drug development is gradually shifting toward genuine original research and innovation, advancing toward “global first-in-class” breakthroughs. Small-molecule innovative drugs remain the mainstay of pharmaceutical development. The environment for China’s small-molecule innovative drug industry is improving, with new products continually emerging; however, significant challenges persist, including insufficient innovation capacity, inadequate R&D investment, limited market potential for products, and rudimentary capital operations. Future industry development will be further guided by clinical needs, driven by source innovation, anchored in the domestic large-scale circulation as the main body, and propelled by the dual-circulation strategy integrating domestic and international markets. By targeting the global market and benchmarking against first-in-class novel drugs, the industry will strive to achieve robust growth and prosperity.

Where Is the Technological Tipping Point for Winning in the Small-Molecule Drug Market?

In recent years, the rise of novel biologics such as antibodies, gene therapies, and cell therapies has strongly captured a portion of the small-molecule drug market. Meanwhile, small-molecule drug development has been constrained by the slowing annual growth in the discovery of new targets and the diversity of small-molecule libraries, resulting in relatively low success rates. However, advances in emerging technologies—including PROTACs, molecular glues, and AI-driven drug discovery—have introduced new R&D paradigms, reinvigorating small-molecule drug development. Currently, numerous companies in China are strategically positioning themselves in this field.

Why Do Emerging Small-Molecule Degraders Hold Immense Potential?

Small-molecule degraders have gained significant momentum in new drug development in recent years, owing to their unique catalytic mechanisms that enable targeting of traditionally “undruggable” targets and address critical challenges such as drug resistance. In 2021 alone, incomplete statistics indicate that 13 financing rounds were directed toward this niche sector in China, raising over RMB 1 billion. Internationally, U.S. pharmaceutical companies Arvinas and C4 Therapeutics are leading the development of small-molecule degrader drugs, with their respective pipelines having advanced to Phase II clinical trials.

Although no small-molecule degrader drugs have yet been commercially launched, clinical pipelines currently in development demonstrate that these agents exhibit excellent safety and efficacy profiles, holding broad therapeutic potential across multiple indications, including cancer and autoimmune diseases.
