February 28, 2019 News /Bio ValleyBIOON/ -- U.S. pharmaceutical giantPfizer(Pfizer) recently announced that new serotypes have been incorporated into its 20-valent pneumococcal conjugate vaccine (20vPnC) candidate, PF-06482077, which is under investigation for the prevention of invasive disease and pneumonia caused by the pneumococcal serotypes included in the vaccine among adults aged 18 years and older.

PF-06482077 includes the 13 serotypes contained in Prevnar 13 (1, 3, 4, 5, 6A, 6B, 7F, 9V, 14, 18C, 19A, 19F, and 23F) as well as seven additional serotypes (8, 10A, 11A, 12F, 15BC, 22F, and 33F).

Pfizer also announced that data from a Phase II proof-of-concept study evaluating 20vPnC in adults had been accepted for an oral presentation at the 29th European Congress of Clinical Microbiology and Infectious Diseases (ECCMID), held in Amsterdam, the Netherlands, from April 13 to 16, 2019. Furthermore, Pfizer has initiated three Phase III clinical trials (NCT03828617, NCT03835975, NCT0376146) to assess 20vPnC in adults. These three clinical trials will enroll more than 6,000 adult participants, including both pneumococcal vaccine-naïve adults and those previously vaccinated against pneumococcus.

All seven new serotypes included in the 20-valent pneumococcal conjugate vaccine (20vPnC) are global causes of invasive pneumococcal disease, and six of these seven serotypes (8, 10A, 11A, 15BC, 22F, and 33F) are associated with high case fatality rates. Furthermore, four of these serotypes (11A, 15BC, 22F, and 33F) are associated withAntibioticsassociated with antibiotic resistance and/or meningitis (serotypes 10A, 15BC, 22F, and 33F). In summary, the 20 serotypes included in the 20-valent pneumococcal conjugate vaccine (20vPnC) collectively account for the majority of invasive pneumococcal disease currently circulating among adults in the United States and globally.

Kathrin U. Jansen, Senior Vice President and Head of Vaccine Research and Development at Pfizer Inc., stated, “The changes in the prevalence of pneumococcal serotypes observed globally are partly due toAntibioticsAntimicrobial resistance has made pneumococcal disease burden remain a significant unmet medical need across all age groups. Over time, we have carefully monitored and assessed global pneumococcal epidemiology and selected additional serotypes for the 20-valent pneumococcal conjugate vaccine (20vPnC) candidate, with the aim—assuming successful development—of expanding global protection against pneumococcal disease beyond the coverage provided by existing pneumococcal conjugate vaccines.(Bioon.com)
