Regeneron and CytomX Expand Collaboration with Up to $4 Billion Deal to Advance Conditionally Activated Bispecific Antibody Therapies

On June 3, 2026, CytomX Therapeutics announced the expansion of its collaboration and license agreement with Regeneron Pharmaceuticals. The two companies will jointly develop conditionally activated bispecific cancer therapies by leveraging CytomX’s Probody® therapeutic platform and Regeneron’s Veloci-Bi® bispecific antibody development platform.

This collaboration commenced on November 17, 2022. According to the agreement at that time,CytomX will receive a $30 million upfront payment and is eligible for future target nomination payments, as well as up to $2 billion in preclinical, clinical, and commercial milestones.CytomX is also eligible to receive tiered global net sales royalties.
Under the expanded agreement, Regeneron and CytomX will continue to collaborate on discovery activities to identify and validate conditionally activated bispecific antibodies. Regeneron will be responsible for funding preclinical and clinical development as well as commercialization activities. CytomX will receive$37 milliontarget nomination fees for the two other selected targets, Regeneron has the right to select up to6 unitsAdditional Future Targets.The total potential nomination payments for expanded collaboration targets, along with preclinical, clinical, regulatory, and commercial milestones, could reach approximately $4 billion.. CytomX is also eligible to receive tiered global net sales royalties on collaborative products.
The key feature of CytomX’s Probody therapeutic platform is that the target-binding sites of therapeutics developed using this platform are masked, thereby avoiding toxicity to healthy tissues. However, due to the high concentration of proteases in the tumor microenvironment, these proteases can unmask the previously concealed target-binding sites once the drug reaches the tumor site, activating the drug and enabling it to kill cancer cells. Consequently, such drugs offer a wider therapeutic window and hold potential for combination therapy with many other agents. Many cancer combination therapies fail because the drug combinations exert excessive toxicity on healthy tissues.

Veloci-Bi is Regeneron’s proprietary bispecific antibody platform. This technology platform enables the generation of full-length bispecific antibodies that resemble natural antibodies, allowing for manufacturing using standard antibody production technologies and exhibiting pharmacokinetic properties similar to those of conventional antibodies.
