Recently, Kite, a Gilead Sciences company, announced that its CAR-T therapy Yescarta achieved positive results in a Phase 2 clinical trial as a first-line treatment for patients with high-risk large B-cell lymphoma (LBCL). Following a single infusion of Yescarta, 85% of patients (n=27) achieved clinical response, including 74% who attained complete response. The press release noted that these represent the first positive clinical results for a CAR-T therapy used as a first-line treatment.
Yescarta is the second FDA-approved CAR-T therapy, having received approval as a third-line treatment for patients with refractory large B-cell lymphoma (LBCL). It works by equipping T cells harvested from the patient with chimeric antigen receptors (CARs) targeting CD19, thereby transforming these T cells into a cell therapy capable of killing tumor cells.
In the multicenter, open-label, single-arm Phase 2 clinical trial named ZUMA-12, Yescarta achieved a complete response rate of 74% at a minimum follow-up duration of one month. Furthermore, at a median follow-up time of 9.3 months, 70% of patients who had achieved a response remained in response. At a median follow-up time of 9.5 months, the median progression-free survival, overall survival, and duration of response had not yet been reached.
