“Recurrent Glioblastoma = Countdown to Life” – This “death curse” plaguing brain tumor patients worldwide was finally shattered in early 2026 by a novel NK cell combination therapy!

As the most malignant primary brain tumor in adults, glioblastoma (GBM) has long been synonymous with devastation for patients: even after undergoing "maximal therapy" comprising surgery, radiotherapy, and temozolomide chemotherapy, the recurrence rate remains close to 100%, with a median survival of only 6–9 months post-recurrence and a 5-year survival rate of less than 10%...

But in January 2026, the release of data from the QUILT-3.078 trial by U.S.-based ImmunityBio, Inc. sent shockwaves through the entire brain tumor field — a triple-combination therapy centered on natural killer (NK) cells not only resulted in a median overall survival that was “not yet reached” in patients with recurrent glioblastoma (Far Exceeding the Historical Upper Limit), with a favorable safety profile, it even reversed the immune collapse caused by radiotherapy and chemotherapy. The “glimmer of hope” for survival in brain tumor patients has truly arrived!

Breaking the 6–9-Month Deadlock! NK Cell Triple Therapy Doubles Survival in Patients with Recurrent Glioblastoma

Traditional therapies either “destroy immunity” (chemotherapy depletes lymphocytes) or “fail to penetrate” (drugs cannot cross the blood-brain barrier), whereas this innovative triple-combination regimen directly employs a “multi-pronged approach.”The trial, codenamed QUILT-3.078 (NCT06061809), also hailed as a “milestone” in brain tumor therapy, addresses three major therapeutic impasses in glioblastoma treatment through a powerful triple-pronged approach characterized by “chemotherapy-free, potent immunotherapy, and high safety.”

I. Mechanism Innovation: 1+1+1>3, Precisely Overcoming “Immunosuppression + Blood-Brain Barrier”

ANKTIVA® (nogapendekin alfa)

IL-15 receptor superagonist, like an “immune power bank”—activates only NK cells and CD8+ T cells without activating immunosuppressive Treg cells, and can repair immune damage caused by radiotherapy and chemotherapy, ensuring that anticancer cells “only increase, never decrease”;

PD-L1 t-haNK

Genetically engineered “smart NK cells” are equipped with surface-bound PD-L1 antibodies for targeted navigation, enabling precise locking onto brain tumor cells. These cells penetrate the tumor microenvironment with threefold greater efficiency than conventional NK cells and leverage a newly discovered “command mechanism” reported in Cell—secreting CCL5/XCL1 chemokines to recruit dendritic cells and activate T cells, thereby launching a “global immune attack.”

Bevacizumab and Tumor Treating Fields

Anti-angiogenic “starvation agents” not only cut off tumor nutrient supply but also open up “channels” in the blood-brain barrier, allowing NK cells to smoothly enter brain lesions and resolve the longstanding problem of “drugs being unable to penetrate.”

“This is the first time that ‘immune reconstitution + precise cancer killing + microenvironment remodeling’ have been combined to target all weaknesses of glioblastoma.” Dr. Simon Khagi, research director at the Hoag Family Cancer Institute, emphasized that this combination completely breaks away from the old framework of “aggressive chemotherapy.”

II. Breakthrough Data: 19 Out of 23 Enrolled Patients Survived, Reversal of Immune Collapse

As of the data cutoff in January 2026, the trial enrolled 23 patients with recurrent glioblastoma (all belonging to a “refractory population” characterized by prior failure of radiochemotherapy and severe lymphopenia), delivering results that have rewritten history:

Record-Breaking Survival

Among the 14 patients with evaluable efficacy, the median overall survival (OS) remained “not reached” at a median follow-up of 6 months. Notably, the historical median OS for this patient population peaked at only 9 months. Currently, 19 patients remain alive, and all four deaths occurred within the evaluable cohort; some patients have survived beyond 12 months and are in good condition.

Immune Function and Lymphocyte Recovery

The patient experienced “severe lymphopenia” due to chemoradiotherapy prior to treatment and was enrolled thereafter.Lymphocyte counts recovered and remained stable without chemotherapy. Notably, near-complete responses were observed in broader clinical and compassionate-use experience, with patient survival exceeding 12 months from the date of documented disease progression, which is extremely rare in recurrent glioblastoma.

High Safety Profile:

The treatment regimen demonstrated a favorable safety profile. Notably, no cases of cytokine release syndrome (CRS) or immune effector cell-associated neurotoxicity syndrome (ICANS) were observed.

Even more encouragingly, among the expanded population spanning first- to third-line treatments, all seven patients receiving first-line therapy achieved “complete survival”—suggesting that the NK triple-combination therapy not only rescues “patients with late-stage relapse” but may also serve as a “powerful tool against recurrence” for those in earlier stages.

Domestic Surge in Parallel: NK Cell Therapy Enables Long-Term Survival Across Multiple Cancer Types

Alongside international breakthroughs, NK cell therapy in China is also achieving “precise implementation,” demonstrating remarkable potential in treatment regimens for various solid tumors and bringing renewed hope to patients:

2. Historic Breakthrough in China’s Novel NK Cell Therapy! The World’s First FDA Orphan Drug Designation for Liver Cancer Surges Hope for Survival!

Recently, the PBMC-derived allogeneic NK cell (PANK) therapy, independently developed by a Chinese biopharmaceutical company, successfully obtained Orphan Drug Designation (ODD) from the U.S. Food and Drug Administration (FDA) for the indication of hepatocellular carcinoma (HCC). Notably, this is the first NK cell therapy worldwide to receive FDA ODD for HCC, and this “world’s first” achievement comes from China!

From the international “NK triple therapy” enabling glioblastoma patients to achieve “record-breaking survival,” to the widespread clinical adoption of NK cell therapies in China, malignant tumors once deemed “incurable” now finally offer the possibility of “long-term survival.”

For patients, the most important step now is not to be intimidated by the prognosis of “only six months left,” but rather to: ① undergo relevant tests (PD-L1, ALC) as soon as possible; ② organize medical records and seek evaluation from a professional team,You may submit your medical records to the Medical Department of Global Oncologist Network for assessment of your immune status: 4006667998.③ Proactively embrace new technologies — After all, tumor treatment in 2026 is no longer about “leaving fate to chance,” but rather “taking the initiative to win.”

You could be the next to return to normal life!
