China Securities Journal News (Reporter: Fu Suying) — The China Securities Journal reporter learned from JD Health on September 15 that Xgeva (denosumab injection), a product under BeOne Medicines, recently made its official online debut on JD Pharmacy. With over a decade of clinical application experience worldwide, this medication is China’s first and currently only approved RANKL inhibitor for preventing skeletal-related events (SREs) associated with bone metastases from solid tumors. It was previously approved in China for the treatment of giant cell tumor of bone that is unresectable or where surgical resection may cause severe functional impairment. The drug is expected to provide reliable support for oncology patients in China in the treatment of giant cell tumor of bone and the prevention of SREs related to tumor bone metastases.
It is reported that Xgeva (denosumab) was developed by Amgen, Inc. in the United States, and BeOne Medicines previously secured licensing rights through a global oncology strategic collaboration with Amgen.
Giant cell tumor of bone is a primary borderline bone tumor primarily characterized by osteolytic destruction, with aggressive behavior and metastatic potential. Globally, it accounts for approximately 4%–5% of all primary bone tumors, but is more prevalent in China, where it constitutes about 20% of all primary bone tumors. The tumor predominantly arises in the long bones of the extremities, as well as the axial skeleton and pelvic bones. It is characterized by marked local invasiveness, a high recurrence rate following tumor curettage, and prolonged patient survival.
It is reported that Xgeva (denosumab) exerts a dual effect of inhibiting tumor growth and reducing bone destruction, enabling multidisciplinary collaborative diagnosis and treatment for giant cell tumor of bone (GCTB) at sites where complete surgical resection is difficult and recurrence rates are high, thereby effectively controlling disease progression. Studies have shown that in patients with unresectable disease, Xgeva can achieve long-term disease control, with a 5-year progression-free survival (PFS) rate of 88%; in patients with resectable disease, 80% of subjects receiving neoadjuvant Xgeva therapy showed clinical improvement. Currently, the indication for Xgeva in the treatment of giant cell tumor of bone that is unresectable or where surgical resection may result in severe functional impairment has been included in the 2020 National Reimbursement Drug List.
In addition to giant cell tumor of bone, skeletal-related events (SREs) associated with bone metastases from solid tumors and multiple myeloma also significantly impact patient survival. Without timely preventive treatment, patients face the risk of bone complications that result in impaired mobility and increased pain, which in severe cases may further shorten survival. In November 2020, Xgeva was approved for the prevention of SREs in patients with bone metastases from solid tumors and multiple myeloma, becoming the first and only RANKL inhibitor approved to date for the prevention of SREs.
