Amgen Reports Q3 Earnings: KRAS Inhibitor Lumakras Generates $36M, Total Revenue Reaches $6.7B

On November 2, Amgen announced its third-quarter financial results, reporting total revenue of $6.706 billion for the quarter, up 4% year-over-year. Total revenue for the first nine months reached $19.1 billion.

Sales of the KRAS inhibitor Lumakras/Lumykras reached $36 million this quarter, with cumulative sales totaling $45 million since its market launch on May 28. Over 500 physicians have prescribed Lumakras.

Key developments for Lumakras this quarter include:

  • Approved in Canada and the UK for second-line treatment of NSCLC, currently under regulatory review in the European Union and Japan;
  • At the ESMO 2021 congress, Amgen presented data on its combination with the EGFR monoclonal antibody Vectibix; a Phase III trial for the third-line treatment of mCRC will be initiated in the fourth quarter;
  • At WCLC 2021, Amgen presented Phase I/II NSCLC biomarker and brain metastasis data;
  • At the AACR-NCI-EORTC 2021 conference, Amgen presented data on the combination of Lumakras with trametinib and afatinib;
  • A Phase II study for the first-line treatment of NSCLC will be initiated in patients with STK11-mutant and/or PD-L1-negative tumors.

To date, Amgen has established the most comprehensive global clinical development pipeline targeting the KRAS G12C mutation. This includes combination therapies with anti-PD-L1 monoclonal antibodies, RAF/MEK inhibitors, and EGFR inhibitors for the second-line treatment of NSCLC, as well as combination regimens with PD-1 monoclonal antibodies, SHP2 inhibitors, and mTOR inhibitors for the second-line treatment of NSCLC, CRC, and other solid tumors.

The three autoimmune disease drugs Enbrel (etanercept), Otezla (apremilast), and the adalimumab biosimilar Amgevita collectively contributed $2.009 billion in revenue to Amgen, accounting for 30% of total revenue in the third quarter.

The osteoporosis drug Prolia (denosumab) grew by 15% to reach $803 million, standing as one of Amgen's pillar products. Sales of XGEVA, developed for indications including bone metastases from solid tumors and giant cell tumor of bone, reached $517 million. Driven by the U.S. market, EVENITY, a novel anti-SOST monoclonal antibody for osteoporosis, grew by 152% to reach $149 million.

PCSK9 inhibitor Repatha (evolocumab) generated third-quarter revenue of $272 million (33%). Oncology biosimilars MVASI (bevacizumab) and KANJINTI (trastuzumab) recorded $274 million and $116 million, respectively. Bispecific antibody Blincyto (blinatumomab) demonstrated strong growth (+40%), achieving quarterly sales of $125 million.
