On March 20, the CDE website showed that Sanofi's SAR443122 (eclitasertib, DNL758) was approved for clinical trials, with the indication being ulcerative colitis.

SAR443122 is a selective receptor-interacting serine/threonine-protein kinase 1 (RIPK1) small molecule inhibitor. In October 2018, Sanofi and Denali reached a global development and commercialization collaboration for two RIPK1 inhibitors, SAR443122 and SAR443820 (DNL788).
RIPK1 is a key signaling protein in the tumor necrosis factor (TNF) receptor signaling pathway, regulating inflammation and cell death in tissues throughout the body. Studies have shown that increased RIPK1 activity in the brain drives neuroinflammation and programmed necrosis, and is associated with various central nervous system (CNS) and peripheral diseases.

RIPK1 Inhibitor Entering Phase II Clinical Trials

Currently, no RIPK1 inhibitors have been approved for marketing globally, and there are six products that have entered Phase II clinical trials. Among them, GFH312 from China's Jinfang Pharmaceuticals has also entered Phase II clinical trials, with an indication for intermittent claudication.
