AbbVie Terminates Collaboration on Conditionally Activated CD71 ADC CX-2029

On March 27, CytomX Therapeutics, Inc. announced the termination of its collaboration with AbbVie Inc. regarding CX-2029 (a conditionally activated CD71 ADC) while releasing its financial report for 2022.


CX-2029 is a conditionally activated ADC targeting the transferrin receptor CD71. In January 2023, updated results from the Phase II cohort expansion study targeting squamous esophageal cancer, squamous non-small cell lung cancer (sqNSCLC), and head and neck squamous cell carcinoma (HNSCC) were released. The Phase II study data showed that in unselected patients with squamous tissue tumors who had previously received multiple lines of therapy, CX-2029 demonstrated encouraging clinical activity, with an objective response rate (ORR) of 21% in squamous esophageal cancer patients and an ORR of 10% in sqNSCLC patients.

The incidence of adverse events (AEs) was consistent with the findings observed in Phase I studies, with anemia (all grades: 82.6%, grade 3 or higher: 76.1%) being the most common treatment-related adverse event (TRAE).

On March 21, 2023, AbbVie notified that it would not advance CX-2029 into additional clinical studies and terminated the licensing and collaboration agreement reached in 2016 for this candidate drug.

CytomX stated that it will evaluate the next potential plans for CX-2029 and seek development strategies for a new generation of CD71-targeted therapies.
