AbbVie Discontinues Development of TNFα-Targeted ADC ABBV-154 Due to Unfavorable Benefit-Risk Profile

On April 27, according to foreign media reports, AbbVie announced in its Q1 2023 earnings call that it would terminate the development plan for the TNFα ADC ABBV-154 project.


ABBV-154 consists of adalimumab targeting tumor necrosis factor α (TNFα) and a novel glucocorticoid receptor modulator (GRM). It delivers the glucocorticoid receptor modulator directly to activated immune cells expressing TNFα, aiming to regulate TNF-mediated inflammation-related pathways and reduce systemic inflammatory levels. Previously, AbbVie planned to develop ABBV-154 for indications including rheumatoid arthritis (RA), polymyalgia rheumatica (PMR), and Crohn's disease (CD).

However, AbbVie disclosed in the third quarter of 2022 that although ABBV-154 showed some efficacy in RA patients, it was decided to abandon the development of this indication after comprehensive consideration, focusing on the development of PMR and CD indications.

Regarding the discontinuation of the ABBV-154 project development this time, Thomas Hudson, Senior Vice President and Chief Scientific Officer of AbbVie's R&D Department, stated: "Although ABBV-154 has shown some efficacy, we have also observed changes in biomarkers consistent with higher-dose systemic steroid exposure. The benefit-risk ratio is not sufficient to differentiate ABBV-154 from other available treatments."
