To address one of the biggest challenges in ophthalmic care — the widespread issue of poor patient adherence to treatment — they set a singular goal: "long-term sustained drug delivery." This led to the creation of the SpyGlass drug delivery platform.

SpyGlass Pharma was incubated and established by SpyGlass Ophthalmics. Its parent company, SpyGlass Ophthalmics, is a private ophthalmic treatment company that develops ophthalmic devices aimed at improving patients' vision and eye care. Its devices address unmet needs in cataracts, lens replacement, surgeries, and other chronic ophthalmic conditions, enabling patients to access high-quality ophthalmic treatments at an affordable cost. SpyGlass Ophthalmics founded its subsidiary, SpyGlass Pharma, with the aim of fully leveraging its expertise and resources in the ophthalmology field to explore new market spaces, specifically long-term ophthalmic drug delivery.

 

Malik Y. Kahook stated, "We know that many patients do not adhere to prescribed topical treatments in most cases for a variety of reasons, including adverse reactions such as burning sensation, stinging, blurred vision, inability to correctly administer the eye drops, and forgetfulness." Therefore, the goal was set to "create a drug delivery platform that can provide sustained medication for weeks or even years," which has been translated into practice as the SpyGlass drug delivery platform.

 

The goal of the SpyGlass drug delivery platform is to provide uninterrupted and long-lasting bimatoprost treatment for glaucoma or ocular hypertension patients, which can last for years and reduce intraocular pressure (IOP). Bimatoprost is highly safe and effective, and all known safety information about this active drug has been well translated into clinical trials. It is considered the most potent topical anti-glaucoma drug currently available for lowering IOP. It is effective for normal individuals, those with POAG (primary open-angle glaucoma), and OHT (ocular hypertension), and is mainly used for POAG or OHT patients who cannot tolerate other IOP-lowering drugs or for whom other IOP-lowering drugs are ineffective. In addition to bimatoprost, the drug-eluting pad of the SpyGlass drug delivery platform is uniquely designed with multifunctionality and may also have the potential to treat various eye diseases, including chronic uveitis and macular degeneration.


Solve the Patient Compliance Challenge


The SpyGlass Drug Delivery Platform is the first and only controlled-release drug delivery platform suitable for all cataract surgeons, as well as the world's first controlled-release drug delivery platform mounted on an IOL (intraocular lens), capable of providing sustained intraocular drug delivery treatment. It consists of a one-piece hydrophobic acrylic IOL and two drug-eluting pads, which can be directly implanted into the capsular bag during routine cataract surgery. These drug-eluting pads can slide onto each haptic of the IOL and are securely fixed at the haptic junctions. Once the drug pads are firmly attached, the IOL and drug-eluting pads are loaded into a standard IOL injector, injected, and implanted into the capsular bag through an incision smaller than 2.4 millimeters. The drug pads remain outside the visual axis, continuously releasing active drugs into the aqueous humor, which then carries the active drugs to the target tissues.

 

As a chronic disease, one of the most crucial aspects of glaucoma care is treatment adherence. Non-adherence to medication may lead to complications, disease progression, and vision loss. Average IOP Reduction of 45% Across All Treatment Groups; Multiple U.S. Clinical Trials to Continue


New Findings Unveiled at Eyecelerator Innovation Conference During the 2022 American Academy of Ophthalmology Annual Meeting in Chicago – Three-Month Data from the First Human Trial of the SpyGlass Drug Delivery Platform Show It Can Reduce IOP in Patients with Open-Angle Glaucoma or OHT. In this human trial, a total of 23 patients undergoing cataract surgery were randomly assigned to use the SpyGlass drug delivery platform and received one of three doses of bimatoprost. After three months, the average IOP reduction across groups was 45%, with all patients achieving an IOP of 18 mmHg or lower without additional treatment. Researchers noted no significant adverse events were reported in the trial, and visual outcomes were similar compared to commercially available intraocular lenses. Newly released six-month data from SpyGlass Pharma’s first-in-human glaucoma treatment trial shows that six months after implantation of the SpyGlass drug delivery platform during cataract surgery, there was an average 45% reduction in IOP across all treatment groups, and 100% of subjects discontinued all topical medications nine months post-implantation.

 

On July 10, 2023, SpyGlass Pharma completed a $90 million Series C financing round to advance new therapies for glaucoma and other chronic ophthalmic diseases. The funding will support multiple U.S. clinical trials for the SpyGlass drug delivery platform. This round was led by RA Capital Management, an investment advisory firm specializing in life sciences and drug development, with participation from Vertex Ventures HC, Vensana Capital, Samsara BioCapital, and New Enterprise Associates. According to Crunchbase, after three rounds of financing, SpyGlass Pharma's total funding has reached $109.8 million. "This financing enables SpyGlass to take a firm step toward Phase 1/2 and Phase 3 clinical trials to support U.S. registration," said Patrick Mooney, CEO of SpyGlass Pharma.

 

The SpyGlass Drug Delivery Platform is not yet FDA-approved for any use. Therefore, Spyglass Pharma is conducting multiple clinical trials in the United States to evaluate the safety and efficacy of its platform in treating glaucoma and other ophthalmic diseases, while also exploring its potential applications in other ophthalmic indications.
