Currently, marketed immune cell therapies such as CAR-T are all based on autologous cell modification, which involves a long process and certain requirements for the patient’s condition. The price, reaching up to a million yuan, greatly limits the popularization of these therapies. All CAR-T products available in and outside China target hematological tumors, while patients with solid tumors account for about 90% of all cancer cases, representing a huge population in urgent need of a breakthrough.

 

Solid tumors have a complex immunosuppressive microenvironment, highly heterogeneous antigen targets, and a physical barrier extracellular matrix, making them difficult to treat. T cells and NK cells primarily operate within the blood and lymphatic circulation, with poor infiltration into solid tumors. As a result, researchers have shifted their focus to other immune cells in the hope of developing new therapies.

 

Macrophages (abbreviated as M cells) are a type of immune cell that easily infiltrates the tumor microenvironment (TME) of solid tumors. They not only directly phagocytose and kill tumor cells but also activate T cells through antigen presentation, stimulate NK cell expansion, and alter the tumor microenvironment by secreting specific cytokines. In recent years, engineered macrophages have become a new direction for the treatment of solid tumors.

Induced Pluripotent Stem Cells (iPSC) refer to a type of pluripotent stem cells with characteristics similar to human embryonic stem cells, which are cultured through the artificial reprogramming and reverse differentiation of somatic cells. iPSCs possess multi-directional differentiation and strong self-replication potential, and under certain conditions, can differentiate into various functional cells. Hundreds of millions or even billions of clinically relevant phenotypic cells, such as cardiomyocytes, neurons, and islet cells, can be cultured in vitro, providing a new pathway for universal cell therapy. In recent years, several regenerative cell and immune cell therapies based on iPSC differentiation have entered clinical stages.

Currently, the two most advanced pipelines are SY001, an autologous monocyte-derived CAR-M therapy, for which an IIT clinical trial has been initiated and is actively recruiting patients, with an expected IND submission in the first quarter of 2024; and SY005, an allogeneic iPSC-derived CAR-M therapy, which is also expected to enter the IND submission stage in 2024. Other pipelines based on different targets and technology platforms are also advancing.

So far, CellOrigin Bio has completed financing amounting to hundreds of millions of RMB.
