Roche Bets Over $2 Billion on Orionis Biosciences’ Molecular Glue Therapies

On September 20, Orionis Biosciences announced a multi-year collaboration with Genentech, a subsidiary of Roche, to jointly develop novel small-molecule drugs targeting challenging targets in major disease areas, including cancer and neurodegenerative diseases.

Orionis' Allo-Glue™ Platform Utilizes a Variety of Unique Approaches to Discover Drug-Like Small Molecules for Disease Targets That Traditional Drug Discovery Methods Still Struggle to Capture. The Platform Integrates a Set of Proprietary Chemical Biology Technologies, Including Bioassays, Computational Analysis, Chemical Libraries, and Automated Processes, for High-Throughput Discovery, Rational Design, and Optimization of Small Molecules That Facilitate or Induce Protein Interactions in Living Cells.

This includes molecular glues, which promote interactions leading to the degradation of targets (via nearly any ligase) or modulate target function through direct or allosteric regulatory mechanisms. Orionis Biosciences' comprehensive approach to molecular glue discovery synergistically integrates both target-centric and ligase-driven drug discovery paradigms.

Under the terms of the agreement, Orionis will be responsible for discovering and optimizing molecular glues targeting designated targets by Genentech, while Genentech will oversee subsequent preclinical, clinical development, regulatory submissions, and commercialization of these small molecules. Orionis will receive an upfront payment of $47 million and is eligible for development milestone payments, as well as potential commercial and net sales milestone payments that could exceed $2 billion, along with tiered royalties on sales of collaboration products.
