
Developer of Novel Immunotherapy
September 27, 2023DayElicio Therapeutics, Inc. announced preliminary relapse-free survival (RFS) data from the ongoing Phase 1 (AMPLIFY-201) study of its lead asset ELI-002. The study evaluates ELI-002 2P (a two-peptide formulation targeting G12D and G12R mutations in KRAS-driven cancers) as a monotherapy for patients with KRAS-driven mutant solid tumors.

"Patients with mutant KRAS cancers, particularly pancreatic and colorectal cancer, face a poor prognosis and limited treatment options when tumor DNA or protein biomarkers are detected after standard surgery and chemotherapy. We are encouraged by the early data showing that T cells induced by ELI-002 can positively impact clinical endpoints, including recurrence and risk of death in this study," said Dr. Eileen M. O'Reilly, Winthrop Rockefeller Professor of Medical Oncology.
Christopher Haqq, MD, Executive Vice President, Head of R&D, and Chief Medical Officer of Elicio, added, "These data further support the mechanism of action of ELI-002 and the potential benefits of lymph node-targeted cancer vaccines. Based on the data presented in this paper and previously reported data at ASCO 2023, we have demonstrated that ELI-002 induces a robust mKRAS-specific T-cell response, including both CD4 and CD8 populations, correlating with tumor biomarker responses, suggesting that this may lead to improved clinical outcomes. We are encouraged by these early observations and look forward to continuing to track and report additional clinical and biomarker data from the AMPLIFY-201 study. Furthermore, the independent Data Monitoring Committee of the AMPLIFY-7P study supports the initiation of a randomized Phase 2 trial investigating ELI-002 7P as a monotherapy for adjuvant treatment in PDAC patients."
About ELI-002
ELI-002 is a novel experimental AMP therapeutic immunotherapy targeting mutations.KRASKRAS mutation-driven cancer. KRAS mutations are one of the most common mutations in human cancers. The seven KRAS-driven mutations targeted by the ELI-002 7P formulation are present in 25% of solid tumors. Specifically, 93% of pancreatic ductal adenocarcinomas and 52% of colorectal cancers (the two most common types in the AMPLIFY-201 study) are positive for KRAS mutations. Additionally, 27% of non-small cell lung cancers are positive for KRAS mutations. ELI-002 consists of AMP-modified mutant KRAS peptide antigens and ELI-004, an AMP-modified immunostimulatory oligonucleotide CpG adjuvant. The AMP mKRAS peptides and AMP CpG target lymph nodes, where they may potentially enhance the function of key immune cells.
