On October 17, Roche and Monte Rosa Therapeutics (referred to as Monte Rosa) reached a strategic collaboration and licensing agreement to discover and develop molecular glue degraders (MGD) targeting previously undruggable cancer and neurological disease targets.

Under the terms of the agreement, Monte Rosa will receive a $50 million upfront payment and is eligible for potential additional payments exceeding $2 billion in preclinical, clinical, commercialization, and sales milestones, as well as tiered royalties.

The two parties also agreed on a mechanism to expand cooperation on multiple targets within the first two years. In this case, Roche will also pay additional nomination, preclinical, clinical, commercialization, and sales milestone payments, as well as tiered royalties on the resulting products.

Monte Rosa will carry out discovery and preclinical research efforts targeting multiple selected cancer and neurological disease targets, while Roche has the option to further advance the preclinical and clinical development of compounds. Monte Rosa retains full rights to its pipeline programs.

Monte Rosa Therapeutics owns a platform for developing molecular glue degraders called QuEEN (Quantitative and Engineered Elimination of Neosubstrates), which can systematically identify therapeutically relevant protein targets suitable for molecular glue degrader development and rationally design molecules while optimizing their potency and selectivity.
