As of December 2022, in the Phase I clinical trial (NCT05287165) evaluating the safety, efficacy, and pharmacokinetic characteristics of IM96 CAR-T therapy for GCC-positive advanced metastatic colorectal cancer (mCRC) that failed ≥3 lines of treatment, a total of 9 patients were enrolled and infused with 3×10.8(DL1)、6×108(DL2)、12×108(DL3) or 20×108(DL4) dose of CAR-T cells. The enrolled patients included 5 males and 4 females, with a median age of 52.6 years.
Clinical research data are as follows:
No neurotoxicity syndrome (ICANS) or cytokine release syndrome (CRS) of grade ≥3 was observed in all enrolled patients.
The disease control rate (DCR) was 66.7%, and the objective response rate (ORR) was 11.1%., one patient achieved partial remission (PR) and has continued to show remission for 11 months to date.;
After CAR-T infusion, among the 9 patients, 5 (55.6%) showed a significant decrease in CEA levels, while all patients had abnormally high CEA levels at baseline. CAR-T cells expanded in all patients and peaked 7 to 10 days after infusion. In 2 patients, the tumors continuously shrank, with CEA levels decreasing within 3 months while CAR-T cells expanded to 10 times.8/L. When the dose of IM96 is ≥6×108,When the moderate to high intensity GCC expression rate in tumor tissue was greater than 30%, all patients achieved tumor shrinkage.
In summary, IM96 in the treatment of ≥3 lines of therapy failure in GCC-positive advanced metastatic colorectal cancer (mCRC)Well-tolerated, with encouraging safety and efficacy, and significant treatment benefits for patients., laying the foundation for further clinical research.
Currently, colorectal cancer is the third most common malignant tumor globally and ranks second among the causes of death from malignant tumors. It has become one of the cancer types that seriously threaten human health.
