November 8, 2023,Eli Lilly (Eli Lilly and Company) announced its glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptor dual agonist Zepbound (tirzepatide) has been approved by the U.S. FDA for weight loss and weight maintenance in obese or overweight adult patients with weight-related comorbidities, including hypertension, dyslipidemia, type 2 diabetes, obstructive sleep apnea, or cardiovascular disease.

Tirzepatide is a once-weekly GLP-1R/GIPR dual agonist that was first approved by the FDA in May 2022 for the treatment of type 2 diabetes.。GIP and GLP-1 can synergistically promote insulin secretion by pancreatic β-cells, exerting a glucose concentration-dependent hypoglycemic effect; additionally, both can improve pancreatic β-cell function and enhance insulin sensitivity; GLP-1 can delay gastric emptying, contributing to an overall hypoglycemic effect.
