On November 15, Generate:Biomedicines, an AI pharmaceutical star company incubated by Flagship, published an article in the Nature magazine about a product namedChroma's Generative AI Model, this model is capable of designing entirely new proteins with excellent biophysical and therapeutic properties that do not exist in nature.

Although protein-based therapies are an important part of the pharmaceutical field, almost all existing therapies focus on modifying proteins found in nature — which only accounts for a small portion of all possible proteins.

Through the Chroma model, Generate:Biomedicines can now harness the power of generative AI to infer generalizable principles governing the relationships between protein sequences, structures, and functions, enabling the rapid design of novel protein molecules with a wide range of therapeutic properties. This will have a significant impact on medicine, materials science, and other fields.

Chroma achieves protein design through Bayesian inference under external constraints, which involve symmetry, substructures, shape, semantics, and even natural language prompts. Experimental characterization of 310 proteins shows that proteins sampled from Chroma are highly expressed, folded, and possess good biophysical properties.

Generate Biomedicines Announces the Release of the Chroma Model to a Wider Research Community (Open Source Code: https://github.com/generatebio/chroma) to Accelerate the Development of Generative Biology.

Generative biology represents a fundamental shift in therapy development driven by generative AI, which goes beyond proteins found in nature to create novel proteins designed to address existing or emerging therapeutic needs.Expected to usher in a brand-new era of programmable drugs,To make drug development faster, cheaper, and more flexible.

Generate Biomedicines CEO Mike Nally stated that models like Chroma are sending a clear signal — biology is becoming programmable.

Generate: Biomedicines is a company founded by Flagship Pioneering. It was established after two years of foundational research in Flagship's lab division and officially "debuted" in September 2020. To date, the company has completed multiple rounds of financing.Total financing of nearly 750 million US dollars, including the C-round financing completed not long ago (September 14) under the capital winter.

In terms of cooperation, in January 2022, Generate:Biomedicines collaborated withAmgenA research collaboration agreement was reached to develop protein therapies for five clinical targets, with a potential transaction value of $1.9 billion. In the same year, the company was also selected for lists such as Fierce 15 and Endpoints 11.

Generate:Biomedicines has built a rich pipeline (nearly 20 projects) involving oncology, immunology, infectious diseases, etc., with the fastest project already entering Phase I clinical trials. The company expects to file multiple IND applications in 2024.

It is worth mentioning that, in addition to Chroma developed by Generate:Biomedicines, on July 11, 2023, David Baker's team from the University of Washington also published a paper in Nature describing a deep learning method called RFdiffusion, which can design completely new proteins from scratch. This method can generate various functional proteins, including topologies never seen in natural proteins ().
