On November 28, Eli Lilly and Company and PRISM BioLab Co., Ltd. reached a licensing and collaboration agreement to develop small molecule inhibitors targeting protein-protein interaction (PPI) sites.


According to the agreement, PRISM will utilize its proprietary PepMetics technology platform to develop small molecule inhibitors for PPI targets selected by Eli Lilly. Eli Lilly has the option to add two more targets within the collaboration and will be responsible for all subsequent clinical development and commercialization efforts. PRISM will receive an undisclosed upfront payment and is eligible for up to $660 million in preclinical, clinical, and commercial development milestone payments, as well as sales royalties.

PepMetics are a class of small molecules that mimic the 3D structures of α-helices and β-turns, which are common peptide structures in intracellular PPI interphase and receptor-ligand interactions. According to PRISM, this technology can transform previously undruggable PPIs into targets that are easily druggable with small molecules, while also enabling the development of oral versions for injectable biologics.

Previously, PRISM also reached collaborations with Roche, Servier, Eisai, and Ohara Pharmaceutical.
