Seismic Therapeutic Secures $121 Million Series B Financing to Advance AI-Driven Immunology Pipeline

Recently, Seismic Therapeutic, Inc., a machine learning immunology company, announced the completion of a $121 million Series B financing round, bringing its total funding to $222 million. The press release noted,This funding will advance the company’s two lead products, S-1117 and S-4321, through clinical proof-of-mechanism, and drive continued growth of its biologics pipeline and expansion of its proprietary IMPACT platform.

Seismic Therapeutic’s proprietary IMPACT platform accelerates the design and analysis of a large number of proteins and protein modifications in parallel by combining machine learning with structural biology, protein engineering, and translational immunology.This parallelized process optimizes biological function and developability simultaneously, creating novel biotherapeutics with superior properties while bypassing the traditional trial-and-error approaches of biotherapeutic development and the challenges of balancing multiple efficacy and manufacturability factors. Seismic Therapeutic, Inc. has an emerging pipeline of biotherapeutics targeting dysregulation of the adaptive immune system to treat autoimmune diseases.

Seismic Therapeutic’s lead products are S-1117, a pan-immunoglobulin (Ig) G protein carving (Sc) enzyme candidate therapy, and S-4321, a PD-1 agonist and Fc γ receptor IIb selective dual-cell bidirectional (DcB) antibody candidate therapy. Since the company’s launch and announcement of Series A financing, Seismic Therapeutic has advanced both programs to the preclinical research stage supporting IND applications. This progress is driven by Seismic’s integration of cutting-edge machine learning technology into drug development within a specific field (immunology biologics). Through this approach and the IMPACT platform, Seismic rapidly designs novel biologics with drug-like properties using multi-parameter optimization techniques.

In the IgSc enzyme project, Seismic Therapeutic, Inc. designed a novel pan-IgG protease,While maintaining enzyme activity and stability, it reduces the immunogenicity and chemical burden of B cells and T cells.Thereby creating a protease therapy that has the potential to address multiple pathogenic mechanisms as a single agent, for the treatment of acute and chronic autoantibody-mediated diseases.Seismic’s DcB Antibody Aims to Optimize Targeting of Multiple Cell Types Driving the Pathogenesis of Autoimmune Diseases, Particularly Through PD-1 Agonism Engaging T Cells and Inhibitory Fc γ Receptor Engagement with Antigen-Presenting Cells.

This Series B financing was led by new investor Bessemer Venture Partners, with additional new investors including Amgen Ventures, Codon Capital, Alexandria Venture Investments, Gaingels, and GC&H. Existing investors Timothy A. Springer, Lightspeed Venture Partners, Polaris Partners, Boxer Capital, GV, Samsara BioCapital, along with management and the founders, also participated. Mr. Andrew Hedin, partner in biotechnology and healthcare at Bessemer Venture Partners, will join the Seismic Therapeutic board of directors.
