Breakthrough News in the "Wolf World" on New Year's Day

China's National Medical Products Administration (NMPA) Center for Drug Evaluation (CDE) announced that DS-7011a, a Class 1 new drug submitted by Daiichi Sankyo (DSKYF.US), has been approved for clinical trials. It is intended to be developed for the treatment of systemic lupus erythematosus (SLE). Public information shows that DS-7011a is an antagonistic antibody targeting TLR7 and is currently undergoing Phase 1b/2 clinical trials overseas for the treatment of SLE and active cutaneous lupus erythematosus (CLE).

It is reported that SLE is a chronic systemic autoimmune disease characterized by multi-system damage symptoms, often leading to multi-organ and multi-system damage, posing a life-threatening condition. The pathogenesis of SLE is complex, involving the proliferation and activation of autoreactive T cells and B cells, the production of various pathogenic autoantibodies, abnormal secretion of cytokines, and abnormal receptor expression.

Studies have shown that TLR7 plays a central role in the pathogenesis of systemic lupus erythematosus (SLE). High expression of the TLR7 gene drives the production of autoantibodies in SLE, and gain-of-function mutations in TLR7 confer susceptibility to SLE. According to preclinical research data published at the 2023 American College of Rheumatology (ACR) meeting, DS-7011a has the ability to inhibit TLR7-stimulated production of inflammatory cytokines and showed no toxicity in monkeys after three months of treatment (including the maximum administered dose). These findings suggest that by targeting and inhibiting TLR7, DS-7011a holds promise as a potential therapeutic option for treating SLE.
