Introduction: SRSD107 is a novel siRNA therapy

Shanghai, China and San Diego, USA, February 2, 2024: Sirius Therapeutics (SHANGHAI) Co., Ltd. ("Sirius Therapeutics"), announced that the first subject dosing has been successfully completed on January 30, 2024, for its self-developed next-generation anticoagulant drug SRSD107. SRSD107 is a novel siRNA therapy for the prevention or treatment of thromboembolic diseases, representing one of the key product pipelines in the company’s cardiovascular disease portfolio. This Phase 1 clinical study mainly evaluates the tolerability, pharmacokinetics, and pharmacodynamics of SRSD107 under single ascending dose and multiple ascending dose conditions.

Dr. Qunsheng Ji, CEO of Sirius Therapeutics, stated: "The dosing of the first subject marks the beginning of our global development of SRSD107 to evaluate its potential to transform traditional anticoagulant drugs, with the possibility of achieving a dosing frequency of six months or longer while reducing the risk of bleeding. Additionally, SRSD101, another siRNA therapy developed by Sirius Therapeutics for treating dyslipidemia, has received IND approval from the National Medical Products Administration in China and has entered clinical trials."

About Thromboembolism

It can occur in either the arterial or venous circulation and is the common pathological basis for most myocardial infarctions, ischemic strokes, and venous thromboembolism (VTE). One in four people worldwide die from diseases caused by thromboembolism.

About SRSD107 Injection

SRSD107 Injection is a double-stranded small interfering RNA (siRNA) drug independently developed by Sirius Therapeutics with proprietary intellectual property rights. By specifically targeting the liver-directed coagulation factor XI (FXI) mRNA, it inhibits the protein expression of FXI, blocking the activation of the intrinsic coagulation pathway, thereby exerting anticoagulant/antithrombotic effects. Preclinical trial data show that a single subcutaneous injection of SRSD107 can achieve nearly 100% knockdown of FXI expression, lasting up to half a year, without any bleeding observed. With its potent, long-lasting efficacy and favorable safety profile, SRSD107 has the potential to become a first-in-class and best-in-class next-generation safer anticoagulant.
