Takeda today announced itsPotential "best-in-class"Results from the Phase 2 clinical trial of the investigational monoclonal antibody mezagitamab (TAK-079) in patients with persistent or chronic primary immune thrombocytopenia (ITP). Analysis shows,The drug demonstrated positive efficacy and safety. Therefore, Takeda plans to initiate the relevant global Phase 3 trial in the 2024 fiscal year.The company expects to have a total of 5 New Molecular Entities (NMEs) in Phase 3 trials during the 2024 fiscal year.

The TAK-079-1004 trial, which has been announced this time, is an ongoing randomized, double-blind, placebo-controlled Phase 2 trial. It aims to evaluate the efficacy and safety of three different doses of subcutaneous mezagitamab administered weekly compared with placebo for the treatment of chronic (duration over one year) or persistent (duration 3-12 months) primary ITP patients, with dosing lasting for eight weeks.
The interim analysis showed that mezagitamab has positive safety and efficacy. Overall, mezagitamab demonstrated good safety and tolerability across all three cohorts. All tested doses of the drug showed a higher platelet response rate than placebo. Additionally,The increase in patients' platelet counts is dose-dependent, with the maximum platelet response observed at the highest tested dose. Moreover, patients treated with mezagitamab rapidly generate a platelet response, which is continuously maintained after treatment.

Mezagitamab is a fully human IgG1 monoclonal antibody that exhibits high affinity for cells expressing CD38, including plasmablasts, plasma cells, and natural killer cells., thereby leading to the exhaustion of these cells.
