Recently, information on ClinicalTrials.gov showed,MSD and Moderna Plan to Launch a Phase 2/3 Clinical Trial to Evaluate the Efficacy and Safety of the Personalized Neoantigen Vaccine mRNA-4157 (V940) in Combination with MSD’s PD-1 Inhibitor Keytruda for the Treatment of Patients with Resectable Locally Advanced Cutaneous Squamous Cell Carcinoma.This is the third pivotal clinical trial planned after the two companies launched pivotal clinical trials using mRNA cancer vaccines to treat melanoma and non-small cell lung cancer.

Moderna’s mRNA vaccine technology can integrate sequences encoding multiple neoantigens into a single synthetic mRNA molecule. The mRNA-4157 cancer vaccine, co-developed by Moderna and MSD, can encode up to 34 neoantigens.It has already demonstrated remarkable efficacy in Phase 2b clinical trials for the treatment of high-risk melanoma patients. Released in December 2023.3-Year Follow-Up ResultsDisplay,Compared with Keytruda monotherapy, the combination of mRNA-4157 and Keytruda significantly improved relapse-free survival (RFS), reducing the risk of recurrence or death by 49% (HR=0.510, 95% CI: 0.288-0.906; one-sided p=0.0095), and improved distant metastasis-free survival (DMFS), reducing the risk of distant metastasis or death by 62% (HR=0.384, 95% CI: 0.172-0.858; one-sided p=0.0077).
